藤梨芪瞿方含药血清对人卵巢癌SKOV-3细胞形态及增殖活性的影响

摘　　要：目的探讨藤梨芪瞿方含药血清对体外培养人卵巢癌SKOV-3细胞形态、增殖活性的影响及可能的机制研究。方法选取SPF级雄性大鼠48只,随机分为生理盐水组、顺铂组、藤梨芪瞿方组及藤梨芪瞿方联合顺铂组,每组各12只。大鼠连续灌胃5 d后,心脏取血。将SKOV-3细胞分为对照组与实验组,实验组分组对应含药血清分组,干预24、48 h后,用于检测。分别采用MTT法检测5%、10%、20%浓度的藤梨芪瞿方含药血清对人卵巢癌SKOV-3细胞增值活性的影响;采用HE染色法观察20%浓度的藤梨芪瞿方对人卵巢癌SKOV-3细胞形态变化的影响。Western blot检测细胞色素C(Cyt C)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)蛋白表达情况。结果藤梨芪瞿方可抑制SKOV-3细胞的增殖活性,并且与含药血清浓度呈正相关;藤梨芪瞿方含药血清处理后的SKOV-3细胞出现凋亡特征性变化;藤梨芪瞿方含药血清处理后的SKOV-3细胞Cyt C、Caspase-3蛋白表达上升。结论藤梨芪瞿方含药血清可抑制人卵巢癌SKOV-3细胞的增殖,促进SKOV-3细胞的凋亡。Objective To study the effect and mechanism of Teng Liqiqu-Fang(TQ group)on the proliferation and apoptosis of human ovarian cancer SKOV-3 cells.Methods Forty eight SPF male rats were randomly divided into normal saline group,cisplatin group,TQ group and TQ combined with cisplatin group,with 12 rats in each group.After continuous intragastric administration for 5 days,blood was taken from the heart.SKOV-3 cells were divided into blank group and experimental group.The experimental group was divided into groups corresponding to drug containing serum.After 24 and 48 hours of intervention,SKOV-3 cells were used for detection.MTT assay was used to detect the effects of 5%,10%and 20%TQ group containing serum on the proliferation activity of human ovarian cancer SKOV-3 cells;HE was used to observe the effect of 20%TQ group on the morphological changes of human ovarian cancer SKOV-3 cells.The expressions of Cyt C and Caspase-3 proteins were detected by Western blot.Results TQ group could inhibit the proliferation of SKOV-3 cells and was positively correlated with the concentration of drug containing serum;There were characteristic changes of apoptosis in SKOV-3 cells treated with TQ group containing serum.The expression of Cyt C and Caspase-3 protein increased in SKOV-3 cells treated with TQ group containing serum.Conclusion TQ group can inhibit the proliferation of human ovarian cancer SKOV-3 cells and promote the apoptosis of SKOV-3 cells.
