基于网络药理学分析公英解毒洗剂溻渍治疗下肢静脉性溃疡的分子机制

摘　　要：目的基于网络药理学分析公英解毒洗剂溻渍治疗下肢静脉性溃疡的分子机制。方法通过TCMSP、BATMAN-TCM数据库检索公英解毒洗剂中牡丹皮、蒲公英、金银花、连翘、甘草、赤芍、白芷、木鳖子、黄柏、苦参的活性成分,并利用UniProt数据库获取公英解毒洗剂的预测靶点。运用DisGeNET、OMIM数据库获取下肢静脉性溃疡的疾病靶点。利用String数据库及Cytoscape 3.7.2获取蛋白质相互作用网络图。通过Metascape数据库进行基因本体(GO)分析及京都基因与基因组百科全书(KEGG)通路富集分析。结果公英解毒洗剂溻渍治疗下肢静脉性溃疡的关键活性成分为槲皮素、胆碱、汉黄芩素、黄连素、山奈酚等,核心靶点涉及血管内皮生长因子A(VEGFA)、白细胞介素-1β(IL-1β)、细胞肿瘤抗原p53(TP53)等。GO分析及KEGG富集分析通路得到54个,主要涉及晚期糖基化终末产物-晚期糖基化终末产物受体(AGE-RAGE)信号通路、癌症信号通路、丝裂原活化蛋白激酶信号通路等。结论公英解毒洗剂通过多成分、多靶点、多通路发挥治疗下肢静脉性溃疡,通过本研究为公英解毒洗剂溻渍治疗下肢静脉性溃疡提供理论支持。Objective To explore the molecular mechanism of gongying jiedu lotion in external medicinal liquid treatment of venous leg ulcer based on the network pharmacology.Method Active ingredients of peony cortex,dandelion,honeysuckle,forsythia,licorice,red peony root,angelica dahurica,mussel,phellodendron,and sophora flavescens in gongying jiedu lotion were identified with the help of TCMSP and BATMAN-TCM database;the UniProt database was used to obtain the predictive target of gongying jiedu lotion;the disease targets of venous leg ulcer were obtained by using DisGeNET and OMIM databases;protein interaction network diagram was obtained by using String database and Cytoscape 3.7.2 gene ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed through Metascape database.Result The key active ingredients of gongying jiedu lotion in the treatment of venous leg ulcer were quercetin,choline,wogonin,berberine,kaempferol,etc.The core targets involved vascular endothelial growth factor A(VEGFA),interleukin-1β(IL-1β),cellular tumor antigen p53(TP53),etc.Fifty-four GO analysis and KEGG enrichment analysis pathways were obtained,which mainly involved advanced glycosylation end products-receptor for advanced glycation end(AGE-RAGE)products signaling pathway,cancer signaling pathway,mitogen-activated protein kinase signaling pathway,etc.Conclusion Gongying jiedu lotion can take effect in treating venous leg ulcer through multiple components,multiple targets and multiple pathways.This study provides theoretical support for gongying jiedu lotion in the treatment of venous leg ulcer.
