摘  要:目的探讨柠檬桉油自微乳递送系统(SMEDDS)的制备及其药剂学性能。方法通过改变处方配比,制备不同的柠檬桉油自微乳,绘制伪三元相图确定微乳液的有效自乳化区域;以自微乳平均粒径为响应值,采用星点设计-效应面法对柠檬桉油自微乳处方进行优化,筛选柠檬桉油自微乳处方中的乳化剂和助乳化剂并验证;取最佳处方制备所得的柠檬桉油自微乳超纯水稀释10倍,观察外观,采用马尔文激光电位粒度仪测粒径大小、分布情况及Zeta电位。结果柠檬桉油SMEDDS的最佳处方是柠檬桉油(45%)为油相、巴斯夫15-羟基硬脂酸聚乙二醇酯(HS15,36.7%)为乳化剂、无水乙醇(18.3%)为助乳化剂、乳化剂及助乳化剂比例为2∶1,滴加纯水9 mL,得外观呈半透明带蓝色乳光的柠檬桉油SMEDDS,激光粒径电位仪测得平均粒径为(49.05±0.33)nm,Zeta电位为(4.66±0.18)mV。结论柠檬桉油SMEDDS制备方法简便,重复性好,所得柠檬桉油SMEDDS外观良好,理化性质稳定。Objective To study the preparation and pharmacy properties of lemon eucalyptus oil self-microemulsion drug delivery system(SMEDDS).Method Different lemon eucalyptus oil self-microemulsion was prepared by changing the prescription ratio to determine the effective self-emulsification region of the microemulsion.Using the average particle size of self-micromilk as the response value,the star point design-effect surface method was optimized,and the emulsifier in the micromilk prescription were screened and validated.The lemon eucalyptus oil obtained from optimal prescription preparation was diluted 10 times from microemulsion ultra-pure water,and the appearance was observed.The particle size,distribution,and Zeta potential were measured by Malvin laser potential granometer.Result The optimal formula for SMEDDS of lemon eucalyptus oil was as follows:lemon eucalyptus oil(45%)as oil phase,HS15(36.7%)as emulsifier,anhydrous ethanol(18.3%)as co-emulsifier,and the ratio of emulsifier to co-emulsifier(Km)was 2∶1.After adding 9 mL pure water,the SMEDDS of lemon eucalyptus oil with a translucent blue opalescence was obtained.The results showed that the average particle size was(49.05±0.33)nm and the Zeta potential was(4.66±0.18)mV.Conclusion The preparation method of lemon eucalyptus oil SMEDDS is simple and reproducible,and has stable physical and chemical properties.
