摘 要:目的建立乌蛇止痒丸中蛇床子素、苍术素和盐酸小檗碱的UPLC-DAD法。方法 Waters Acquity Beh C18色谱柱(100 mm×2.1 mm,1.7μm);检测器:DAD检测器;流动相为乙腈–0.1%磷酸溶液,梯度洗脱;检测波长:331 nm;体积流量:1.0 mL/min;柱温:30℃;进样体积:25μL。结果盐酸小檗碱、苍术素和蛇床子素分别在8.954~447.700、3.980~199.000、8.670~433.500μg/mL具有良好的线性(r≥0.995);盐酸小檗碱、苍术素、蛇床子素平均回收率分别为99.69%、98.78%、99.38%,RSD值分别为1.9%、1.6%、1.5%。结论本法具有快速、准确、简便等特点,为进一步提高乌蛇止痒丸的质量控制提供技术参考。Objective To explore a method for determination of osthol, atractylodin, and berberine hydrochloride in Wushe Zhiyang Pills by UPLC. Methods The HPLC method was carried out on Waters Acquity Beh C18 column(100 mm ×2.1 mm,1.7 μm). Detector was selected as DAD. The mobile phase was consisted of acetontrile-0.1% phosphoric acid solution with gradient elution. The detection wavelength was set at 331 nm. The flow rate was 1.0 mL/min, temperature of column was set at 30 ℃, and the volume of injection was 25 μL. Results Berberine hydrochloride, atractylodin, and osthole showed good linear relationships in concentration range 8.954—447.700 μg/mL, 3.980—199.000 μg/mL, and 8.670—433.500 μg/mL(r ≥ 0.995). Average recoveries of samples were 99.69%, 98.78%, and 99.38% with RSD values as 1.9%, 1.6%, and 1.5%, respectively. Conclusion This method is fast, accurate,and simple, which provides a technical reference for improving the quality control of Wushe Zhiyang Pills.
