摘 要:目的优选鸡蛋花茎皮总黄酮的最佳提取工艺。方法以总黄酮含量为评价指标,在单因素试验基础上,以乙醇体积分数、提取时间、提取次数和提取温度为考察因素,采用L9(34)正交试验法优选最佳提取工艺;以乙醇体积分数、提取时间、提取温度为考察因素,采用星点设计-响应面法优选最佳提取工艺,并进行验证。结果正交试验法所得最佳提取工艺为70%乙醇提取3次,每次90 min,提取温度为60℃,所得总黄酮平均含量为2.43%(RSD=3.36%)。星点设计-响应面法所得最佳提取工艺为75.77%乙醇提取3次,每次77.32 min,提取温度为63.90℃。总黄酮平均含量为1.88%(RSD=1.21%),与预测值(1.89%)相当。结论两种试验方法优化得到的最佳提取工艺相近,星点设计-响应面法优化所得工艺更准确,设计更合理,可用于鸡蛋花茎皮总黄酮的提取。Objective To optimize the extraction process of total flavonoids from Plumeriae Rhizoma et Cortex.Methods On the basis of single factor test,the extraction process was optimized by the L;(3;) orthogonal test with the content of total flavonoids as the evaluation index,and with the ethanol volume fraction,extraction time,extraction times and extraction temperature as the investigation factors.The extraction process was optimized by the central composite design-response surface methodology with ethanol volume fraction,extraction time and extraction temperature as the investigation factors,and the verification test was conducted.Results The optimal extraction process by the orthogonal test was as follows:70% ethanol was used for extraction for three times,90 min each time,the extraction temperature was 60℃,and the average content of total flavonoids was 2.43%(RSD=3.36%).The optimal extraction process by the central composite design-response surface methodology was as follows:75.77% ethanol was used for extraction for three times,77.32 min each time,the extraction temperature was 63.90℃,and the average content of total flavonoids was 1.88%(RSD=1.21%),which was equivalent to the predicted value(1.89%).Conclusion The optimal extraction processes optimized by the two methods are similar,and the process optimized by central composite designresponse surface methodology is more accurate and reasonable,which can be used for the extraction of total flavonoids from Plumeriae Rhizoma et Cortex.
