蟾酥乳膏透皮吸收及促进伤口愈合作用的初步研究

摘　　要：目的:将制备的蟾酥乳膏剂进行体外透皮吸收实验并评价其对皮肤伤口愈合的效果。方法:利用体外Franz扩散池法测定蟾酥乳膏的体外透皮吸收率;采用小鼠断尾法测定小鼠止血时间及出血量;根据全层皮肤缺损法制备兔皮肤缺损模型,考察乳膏对兔皮肤伤口愈合的影响。结果:实验结果表明,制备乳膏剂光泽度、细腻度、延展性良好,具有良好的稳定性,透皮吸收率高,并能缩短小鼠断尾止血时间、减少出血量,并且可以促进家兔皮肤伤口愈合(P<0.05)。结论:本实验确定了蟾酥乳膏剂的体外透皮吸收量,并进行了初步的药效试验。家兔皮肤伤口愈合实验结果:蟾酥乳膏在给药1周后表现出促进伤口愈合的效果,且涂抹乳膏后未见兔皮肤出现红斑、红肿,显示了蟾酥乳膏具有一定的安全性。Objective:To valuate the effect of Bufonis Venenum cream on skin wound healing through in vitro transdermal absorption experiment.Methods:In vitro transdermal absorption rate of Bufonis Venenum cream was determined by Franz diffusion cell.Hemostasis time and bleeding volume were determined by tail-breaking method in mice.Rabbit skin defect model was prepared according to the full-thickness skin defect method to investigate the effect of the cream on skin wound healing.Results:The results showed that the prepared cream performs well on the test in gloss,fineness,ductility,stability and transdermal absorption.It could shorten the bleeding time of tail-breaking,reduce the amount of bleeding in mice and promote skin wound healing in rabbits(P<0.05).Conclusion:The in vitro transdermal absorption of Bufonis Venenum cream was determined and its preliminary efficacy test was carried out.The wound healing test in rabbits showed that Bufonis Venenum could promote the would healing,there was not any signs of erythema and swollen,which manifests the high safety of the cream.
