痛风急性发作期的肠道菌群变化与中医证型的相关性

摘　　要：目的:研究痛风急性发作期的肠道菌群变化及其与中医证型的相关性。方法:收集痛风急性发作患者及健康人群粪便,运用Illumina MiSeq测序技术对各组粪便样本DNA测序。采用Alpha、Beta多样性指数差异比较分析,菌群组成差异分析,菌群LEfSe分析,菌群结构差异热图分析比较各组的差异肠道菌群。结果:痛风急性发作期Alpha多样性降低,差异菌群主要表现为Bacteroidia丰度降低,Negativicutes、Actinobacteria、Bacilli丰度升高。而在不同证型之间,瘀热阻滞组菌属间菌群丰度上升的为Prevotella,丰度降低的为Clostridium;ensu;tricto、Clostridium;lVb、Butyricimonas;湿热蕴结组菌属间菌群丰度上升的为Bacteroides,丰度降低的为Paraprevotella、Dorea、Romboutsia。结论:痛风急性发作期存在肠道菌群变化,不同中医证型存在差异菌群。Objective: To study the changes of intestinal bacteria in acute attack of gout and its correlation with TCM syndrome types. Methods: The stool samples of patients with acute gout attack and healthy people were collected and sequenced by Illumina miseq sequencing technology. Alpha and Beta diversity index difference analysis, flora composition difference analysis, flora lefse analysis and flora structure difference thermogram were used to analyze the intestinal flora of each group. Results: The diversity of Alpha decreased during the acute attack of gout, and the abundance of Bacteroidia decreased, while the abundance of Negativitites, Actinobacteria and Bacilli increased. Among different syndrome types, the abundance of Prevotella flora increased, and the abundance of Clostridium_sensu_stricto, Clostridium_XlVb and Butyrimionas decreased in the blood stasis heat block group. The abundance of Bacteroides intergeneric flora increased and the abundance of Paraprevotella,Dorea and Romboutsia decreased in the damp heat accumulation group. Conclusion: There are changes of intestinal bacteria in acute attack of gout, and there are different flora in different TCM syndrome types.
