摘  要:为分析所收集北苍术种质资源亲缘关系和遗传多样性,从100条ISSR引物中筛选出10个多态性引物进行PCR扩增,利用Popgene 1.32软件计算遗传多样性指数,且运用Ntsys的非加权平均距离法(UPGMA)进行聚类分析,对22个不同来源的北苍术种子个体间亲缘关系和遗传多样性进行研究。结果表明,ISSR试验共得到246个清晰位点,其中224个位点具有多态性,总的多态性比率为91.06%。北苍术种子的观测等位基因数为1.9106±0.2859、有效等位基因数为1.421 7±0.340 9、Nei’s基因多样性指数为0.256 1±0.172 5、Shannon’s信息指数(I)为0.395 9±0.230 6。亲缘关系分析表明,北苍术种质资源的遗传多样性非常丰富,这为其遗传育种、种质资源评估及品种鉴定提供理论基础。In this paper, the genetic relationship and genetic diversity of germplasm resources of Atractylodes chinensis were analyzed.Ten polymorphic primers were selected from 100 ISSR primers for PCR amplification, and the genetic diversity index was calculated by Popgene1.32.In addition, unweighted average distance method(UPGMA) of Ntsys was used for cluster analysis to study the genetic relationship and diversity among 22 A. chinensis seeds from different sources.The results showed that a total of 246 distinct loci were obtained by ISSR analysis, of which 224 loci were polymorphic, with a total polymorphism rate of 91.06%.The observed number of alleles, the effective number of alleles, the Nei’s gene diversity index and Shannon’s information index were 1.910 6±0.285 9,1.421 7±0.340 9,0.256 1±0.172 5,0.395 9±0.230 6 respectively.The genetic diversity of germplasm resources of A. chinensis seeds was very rich, which provided theoretical basis for genetic breeding, germplasm resources evaluation and variety identification.
