摘  要:目的探究基质细胞衍生因子-1(SDF-1)过表达质粒修饰骨髓间充质干细胞(BMSCs)联合丝素/胶原蛋白支架在膝关节软骨缺损动物模型中的修复作用。方法利用蚕丝和胶原蛋白为原材料,构建丝素/胶原蛋白支架,利用SEM显微镜观察其内部结构,并检测支架强度。以新西兰大白兔为研究对象,提取和培养兔源性BMSCs,并且构建SDF-1的过表达载体质粒,通过慢病毒转染BMSCs,通过构建新西兰大白兔的软骨缺损模型,根据是否转染SDF-1过表达质粒分成4组,即模型组、单纯支架组、支架+BMSCs组和支架+SDF-1修饰BMSCs组。利用HE染色和免疫组织化学染色检测软骨缺损的修复作用。结果丝素蛋白/胶原蛋白支架的水平牵拉力和垂直挤压力强度要明显高于单一丝素蛋白支架。支架+SDF-1修饰BMSCs组的修复效果要明显优于模型组、单纯支架组和支架+BMSCs组;支架+SDF-1修饰BMSCs组中II型胶原蛋白的表达量要明显高于模型组、单纯支架组和支架+BMSCs组,说明支架+SDF-1修饰BMSCs组的软骨缺损修复效果好。结论支架+SDF-1修饰软骨缺损模型BMSCs具有更佳的修复效果。Objective To investigate the repairing effect of BMSCs modified with SDF-1 over expression plasmid and fibroin/collagen scaffold on knee joint cartilage defect in New Zealand white rabbit.Methods Silk and collagen were used as raw materials to construct silk fibroin/collagen scaffolds.The internal structure of the scaffolds was observed by SEM,and the strength of the scaffolds was measured.The rabbit bone marrow mesenchymal stem cells(BMSCs)were extracted and cultured,and SDF-1 overexpression vector plasmid was constructed.BMSCs were transfected by lentivirus.The cartilage defect model of New Zealand white rabbit was constructed.The experiments were divided into model group,simple scaffold group,scaffold+BMSCs group and scaffold+SDF-1 repair group.HE staining and immunohistochemical staining were used to detect the repair of cartilage defects.Results The strength of horizontal pulling force and vertical extrusion force was significantly higher in fibroin/collagen scaffolds than in single fibroin scaffolds.The recovery was significantly better in scaffolds+SDF-1 than in model group,scaffolds alone and scaffolds+BMSCs alone.The expression level of type II collagen was significantly higher in scaffolds+SDF-1 than in model group,scaffolds alone and scaffolds+BMSCs alone.Conclusion BMSCs modified by scaffolds+SDF-1 has better repair effect on cartilage defect.
