摘 要:目的探索异氟醚对大鼠局灶性脑缺血再灌注损伤的神经保护机制。方法雄性SD大鼠60只,随机分为假手术组(NC)、缺血再灌注损伤组(IR)、 IR后异氟醚处理组(IRO)、Wnt抑制剂(ICG-001)组(IC)和Wnt激活剂(BML284)组(BM)。根据神经行为学评分判断神经损伤程度,同时通过TUNEL法检测海马区细胞凋亡情况,蛋白印迹实验检测Wnt及β-catenin的表达。结果与IR组相比,IRO组神经行为评分降低;与IRO组相比,Wnt激活剂组神经行为学评分降低,Wnt抑制剂组评分增加(P<0.05)。与IR组相比,IRO组海马区神经凋亡率降低;与IRO组相比,Wnt激活剂组海马区神经凋亡减少,抑制剂组凋亡增加(P<0.05)。与IRO组相比,Wnt激活剂组海马区Wnt/β-catenin蛋白表达增加,而抑制剂组蛋白表达减少(P<0.05)。结论异氟醚可通过Wnt/β-catenin信号通路保护神经细胞,或可成为缺血再灌注的保护药物以阻断神经细胞损伤。Objective To explore the mechanism of neuroprotective effect of isoflurane treatment. Methods Sixty male SD rats were randomly divided into sham operation group(NC), ischemia-reperfusion injury group(IR), isoflurane treatment group after IR(IRO), Wnt signal inhibitor(ICG-001) group(IC) and Wnt signal activator(BML284) group(BM).According to the degree of nerve injury and the behavioral motor ability of the rats, neurobehavioral score was performed to judge the degree of nerve injury. TUNEL assay was used to detect apoptosis in the hippocampal region to determine the protective effect of isoflurane post-treatment and Wnt signal activator or inhibitor. Western blot was used to detect the expression of Wnt and β-catenin to determine the neuroprotective mechanism of isoflurane post-treatment. Results Compared with IRO, neurobehavioral score of Wnt signal activator group decreased, neurobehavioral score of Wnt signal inhibitor group increased(P < 0.05). Compared with the IR group, the hippocampal nerve apoptosis rate was decreased in the IRO group, and decreased in the Wnt activator group and increased in the inhibitor group(P<0.05). Compared with IRO group, Wnt/β-catenin protein expression was increased in Wnt activator group and decreased in inhibitor group(P < 0.05).Conclusion Isoflurane can protect nerve cells by Wnt/β-catenin signal, or it can be used as a protective agent for ischemia reperfusion to block nerve cell injury.
