摘 要:目的检测miR-129-5p在膀胱癌组织和细胞中的表达及其对膀胱癌细胞的作用。方法 RTqPCR法检测miR-129-5p在膀胱癌、癌旁组织以及膀胱癌细胞株中的表达。细胞中过表达miR-129-5p后,CCK-8实验检测细胞增殖能力变化;流式细胞术检测细胞凋亡情况;生物信息学网站预测miR-129-5p的靶点,并用荧光素酶报告基因实验验证,Western blot检测miR-129-5p转染后细胞中靶蛋白的表达情况。结果 RT-qPCR检测发现,miR-129-5p在膀胱癌组织和人膀胱癌细胞系T24和5637中的表达分别低于癌旁组织和人正常膀胱上皮细胞SV-HUC-1细胞。转染miR-129-5p mimics进入T24细胞后,T24细胞增殖受到抑制,凋亡率增加。双荧光素酶报告基因实验证实Wnt5a是miR-129-5p直接作用靶点,Western blot结果证实miR-129-5p能够直接下调Wnt5a的表达。结论 miR-129-5p在膀胱癌组织和细胞中的表达降低,miR-129-5p能够抑制膀胱癌细胞增殖,促进细胞凋亡。Objective To detect the expression of miR-129-5 p in bladder cancer tissues and cells and its effect on bladder cancer cells. Methods RT-qPCR was used to detect the expression of miR-129-5 p in bladder cancer,paracancerous tissues and bladder cancer cell lines. After overexpression of mir-129-5 p in cells, the changes of cell proliferation were detected by CCK-8 experiment. Apoptosis was detected by flow cytometry. The target of mir-129-5 p was predicted by bioinformatics website, verified by luciferase reporter gene experiment, and the expression of target protein of miR-129-5 p transfected cells was detected by Western blot. Results RT-qPCR results showed that the expression of miR-129-5 p in bladder cancer tissue and human bladder cancer cell line T24 and 5637 was lower than that in adjacent tissues and human normal bladder epithelial cells SV-HUC-1 cells. After transfection of miR-129-5 p mimics into T24 cells, the proliferation of T24 cells was inhibited and the apoptosis rate was increased. Double luciferase reporter gene experiment confirmed that Wnt5 a was the direct target of miR-129-5 p, and Western blot confirmed that miR-129-5 p could directly down regulate the expression of Wnt5 a. Conclusion The expression of miR-129-5 p was decreased in bladder cancer tissues and cells, miR-129-5 p could inhibit the proliferation and promote apoptosis of bladder cancer cells.
