摘 要:目的研究两种不同晶面能的纳米硫化镉(NP-CdS)对肺上皮细胞系A549的氧化损伤情况。方法培养A549肺上皮细胞,用不同晶面能的两种纳米硫化镉材料NP-CdS1和NP-CdS2分别处理细胞,染毒24 h后用MTT法检测细胞存活率;用分光光度法检测过氧化氢酶(catalase,CAT)、超氧化物歧化酶(superoxide dismutase,SOD)活力以及丙二醛(malonyldialdehyde,MDA)的含量;采用激光共聚焦显微镜观察细胞内活性氧(reactive oxygen species,ROS)含量的变化;采用电镜技术观察细胞超微结构的变化。结果随着染毒浓度的增加,NP-CdS1和NP-CdS2可引起A549细胞存活率逐渐降低,选择在80%左右存活率的浓度作为后续染毒浓度,即10和20μg/mL;NP-CdS2染毒后细胞内SOD和CAT活力增加,MDA含量增加,且差异均有统计学意义(均P<0.05);NP-CdS1与NP-CdS2染毒后细胞内ROS含量明显升高;电镜下可见硫化镉纳米棒富集在细胞内脂肪滴的周围且排列整齐,并引起细胞核的浓缩。结论纳米硫化镉对肺上皮细胞系A549具有氧化损伤,且晶面能高的纳米材料毒性更强。Objective To study the oxidative damage of cadmium sulfide nanoparticles(NP-Cds)with two different crystal surface energy on A549 pulmonary epithelial cells.Methods A549 pulmonary epithelial cells were cultured and treated with NP-CdS1 and NP-CdS2 respectively,which were NP-Cds with two different crystal surface energy.After exposure for 24 hours,the cell survival rate was measured by the method of MTT.The vitality of catalase(CAT)and superoxide dismutase(SOD),and content of malonyldialdehyde(MDA)were measured by spectrophotometry.The change in content of intracellular reactive oxygen species(ROS)was observed by laser scanning confocal microscope.The changes of cell ultrastructure were observed by using the technique of electron microscopy.Results As the exposure concentration increased,NP-CdS1 and NP-CdS2 could gradually reduce the survival rate of A549 cells.And the concentration resulting in 80%survival rate was chose as the subsequent exposure concentration,that is,10 and 20μg/mL.After NP-CdS2 exposure,the vitality of SOD and CAT,as well as the MDA content increased,and the differences were statistically significant(all P<0.05).After NP-CdS1 and NP-CdS2 exposure,the ROS contents increased significantly.The electron microscope showed that the CdS nanorods concentrated around the fat droplets in the cells and arranged neatly,and caused the nucleus to condense.Conclusion NP-Cds can cause oxidative damage of A549 pulmonary epithelial cells,and the higher the crystal surface energy of nano-materials,the stronger the toxicity.
