摘 要:为了研究钙果试管苗生根基础上的分枝增殖技术,达到一次成苗效果。以钙果4号试管长约1.3 cm的茎尖苗、茎段苗为实验材料,设置不同的生长素种类与水平,研究其一次成苗培养基配方。结果表明,得到茎尖生根率为83%,茎段生根率为65%,茎叶生长良好的基础配方3号:蔗糖20 g·L^(-1)+琼脂粉4.2 g·L^(-1)+IAA 1.0 mg·L^(-1)+MS。仅以茎段苗为实验材料,使用均匀设计U 11(114)对3号配方中的KNO_(3)、NH_(4)NO_(3)、MgSO_(4)·7 H_(2)O、Ca(NO_(3))_(2)·4 H_(2)O进行深度优化,利用回归分析结合Matlab软件求解得到优良配方3-1:蔗糖20 g·L^(-1)+琼脂粉4.2 g·L^(-1)+IAA 1.0 mg·L^(-1)+[Ca(NO_(3))_(2)·4 H_(2)O]340.4 mg·L^(-1)+[NH_(4)NO_(3)]500.2 mg·L^(-1)+[KNO_(3)]9.3 mg·L^(-1)+[MgSO_(4)·7 H_(2)O]567.9 mg·L^(-1)+85 mg·L^(-1)。经验证,该配方茎切段试管苗生根率提升至87.6%,根长为4.28 cm·株^(-1),生根数7.3条·株^(-1),且腋芽萌发成梢明显,嫩梢个数接近4个·株^(-1),嫩梢长度大于2.6 cm·株^(-1),该嫩梢提高了钙果4号瓶内微枝扦插的有效增殖系数,达到了试管苗生根与增殖同步理想的效果。Research the branch propagation technology basing on rooting phase of Cerasus humilis cv.Four in vitro,achieve the one-step-seedling result finally.Taking stem tip and stem segment as experimental materials which reach 1.3 cm longness from Cerasus humilis cv.Four plantlets,set different variety and level of auxin.The results showed that one favorable basic formulation through comparative experiment which labeled 3,including sucrose 20 g·L^(-1)+agar powder 4.2 g·L^(-1)+IAA 1.0 mg·L^(-1)+MS.The root rate of stem tip plantlets inoculated in medium 3 was 83%,the stem segment was 65%comparatively,the leaf and the stem of plantlets grew well in medium 3 totally.Only set stem segment of Cerasus humilis cv.Four plantlets secondly,optimize the concentration of KNO_(3),NH_(4)NO_(3),MgSO_(4)·7 H_(2)O,Ca(NO_(3))_(2)·4 H_(2)O in medium 3 by means of uniform design U 11(114)deeply,apply regression analysis binding Matlab software and compute regression equation results which produced one preferable medium 3-1,including sucrose 20 g·L^(-1)+agar powder 4.2 g·L^(-1)+IAA 1.0 mg·L^(-1)+[Ca(NO_(3))_(2)·4 H_(2)O]340.4 mg·L^(-1)+[NH_(4)NO_(3)]500.2 mg·L^(-1)+[KNO_(3)]9.3 mg·L^(-1)+[MgSO_(4)·7 H_(2)O]567.9 mg·L^(-1)+85 mg·L^(-1).The stem segment of plantlet inoculated in medium 3-1 perform better which compare with medium 3 through repetitive verification,the rooting rate increases to 87.6%,the root length per plantlets was 4.28 cm and the No.of root per plantlets was 7.3.Furthermore,axillary buds germinate and arrive to mature shoot obviously,the No.of axillary shoot got close to 4 per plantlet,the length of axillary shoot exceeds 2.6 cm.These axillary shoots improve the effective proliferation coefficient of Cerasus humilis cv.Four by micro-shoot grafting,attain ideal result which rooting and proliferating carry out simultaneously of Cerasus humilis cv.Four in vitro.
