番鸭源鸭疫里默氏菌大环内脂类药物的耐药基因检测与分析

摘  要:为了解莆田市番鸭源鸭疫里默氏菌对大环内酯类药物的耐药性和相关耐药基因携带情况,对本实验室分离鉴定的49株番鸭源鸭疫里默氏菌菌株采用K-B纸片扩散法进行药敏试验,提取菌株基因组DNA,采用PCR方法对大环内酯类药物的ermA、ermB、ermC、ermF、ereD等5种耐药基因进行检测,阳性产物送公司测序并对序列进行分析。结果表明,49株鸭疫里默氏菌菌株对红霉素、阿奇霉素的耐药率很高,分别为100%(49/49)、89.80%(44/49)。49株对红霉素和44株对阿奇霉素耐药的鸭疫里默氏菌菌株中,均未检测到ermA、ermB、ermC等3种耐药基因,但85.71%以上菌株检测出ermF、ereD两种耐药基因,在鸭疫里默氏菌大环内酯类药物耐药基因中占主导作用,据此推测,目前莆田市鸭疫里默氏菌对大环内酯类药物耐药的主要原因是由ermF和ereD耐药基因介导的。In order to understand the drug resistance of Riemerella anatipestifer isolated from muscovy ducks in Putian city to macrolides and the carrying of related drug resistance genes,49 strains of Riemerella anatipestifer isolated and identified in our laboratory were tested for drug sensitivity by K-B disk diffusion method,and the genomic DNA of the strains was extracted.Five resistance genes including ermA,ermB,ermC,ermF and ereD were detected by PCR,and the positive products were sent to the company for sequencing and sequence analysis.The results showed that 49 strains of Riemerella anatipestifer were resistanct to erythromycin and azithromycin,which were 100%(49/49)and 89.80%(44/49),respectively.ermA,ermB and ermC were not detected in 49 strains of Riemerella anatipestifer resistant to erythromycin and 44 strains resistant to azithromycin,but more than 85.71%of the strains detected ermF and ereD,which played a leading role in the macrolide resistance genes of Riemerella anatipestifer.Therefore,we speculated that the main reason for the resistance of Riemerella anatipestifer to macrolides in Putian city was mediated by ermF and ereD drug resistance genes.
