油茶种子数量相关基因EPFL2的鉴别

摘　　要：拟南芥(Arabidopsis thaliana)AtEPFL2基因属于EPF/EPFL基因家族中的EPFL1/2/3亚家族,与种子数量有关。对普通油茶(Camellia oleifera)幼叶、未成熟种仁、花蕾、根的转录组数据进行挖掘,得到3条油茶EPFL2基因序列;从油茶基因组3代测序数据中鉴别出含有前述3条序列的基因组DNA序列;最后,得到3条油茶EPFL2基因全长CDS序列,分别命名为CoEPFL2a、CoEPFL2b、CoEPFL2c。用MEGA7构建EPFL1/2/3亚家族蛋白质系统进化树,结果表明:CoEPFL2a、CoEPFL2b、CoEPFL2c属于EPFL2分支,并且CoEPFL2a、CoEPFL2c与AtEPFL2在同一个亚分支。用SignalP-5.0预测蛋白信号肽与酶切位点,结果表明:AtEPFL2、CoEPFL2c含信号肽与酶切位点,CoEPFL2a、CoEPFL2b不含;CoEPFL2c可能与AtEPFL2具有同样的功能,即通过调节胚珠密度影响种子数量。AtEPFL2 belongs to EPFL1/2/3 subfamily of EPF/EPFL gene family and is related to seed number in Arabidopsis thaliana.Three CDSs homologous to AtEPFL2 were obtained by mining transcriptome data from Camellia oleifera young leaf,immature kernels,flower buds and roots.Then,3 long DNA fragments containing the CDS were identified from the whole genome SMRT sequence data.Finally,3 full-length CDSs were identified and named CoEPFL2a,CoEPFL2b,CoEPFL2c,respectively.A phylogenetic tree of EPFL1/2/3 subfamily genes was constructed by using MEGA7,which revealed that CoEPFL2a,CoEPFL2b and CoEPFL2c were distributed in EPFL2 clan,and both CoEPFL2a and CoEPFL2c were in a same subclan with AtEPFL2.SignalP-5.0 predicted that there were signal peptides and cleavage sites in both AtEPFL2 and CoEPFL2c,but both CoEPFL2a and CoEPFL2b don′t have.It was suggested that CoEPFL2c might have the same function as AtEPFL2 which affected seed number through regulating ovule density.
