摘 要:目的研究巴戟天地上部分的化学成分。方法采用溶剂萃取、正相硅胶柱、RP-ODS反相柱、Sephadex LH-20凝胶柱和半制备液相等现代柱色谱方法进行系统的分离纯化,通过理化性质及波谱数据分析鉴定化合物的结构,运用CCK8法测定化合物1~5对小鼠单核腹腔巨噬细胞RAW 264.7细胞的细胞毒性并采用NO试剂盒检测其抗炎活性。结果从巴戟天地上部分中分离得到了5个奎尼酸类化合物,分别鉴定为n-butyl 4-trans-O-coumarolquinate(1),hycandinic acid ester-1(2),5-(Z)-coumaroyloxyquinic acid n-butyl ester(3),methyl 4-O-feruloyl-5-O-caffeoylquinate(4)和Hycandinic acid ester-2(5)。化合物1~5对RAW 264.7细胞没有细胞毒作用,其中化合物1、2和4在50μmol·L^(-1)下均具有抑制NO释放的活性。结论化合物1~5均为首次从巴戟天属植物中分离得到,其中化合物1为新的化合物,化合物1、2和4具有潜在的抗炎作用。OBJECTIVE To investigate the chemical constituents from the aerial part of Morinda officinalis How.METHODS Solvent extraction,silica gel column,RP-ODS column,Sephadex LH-20 column and semi-preparative high liquid chromatography and other modern column chromatography methods were used for isolation and purification and structures were identified through physicochemical properties and spectral data analysis.The cytotoxicity and anti-inflammatory activity of compounds 1-5 on RAW 264.7 cells were determined by the CCK8 method and NO method,respectively.RESULTS Five quinic acid compounds were isolated and identified as n-butyl 4-trans-O-coumarolquinate(1),hycandinic acid ester-1(2),5-(Z)-coumaroyloxyquinic acid n-butyl ester(3),methyl 4-O-feruloyl-5-O-caffeoylquinate(4),hycandinic acid ester-2(5).Compounds 1-5 showed no cytotoxicity to RAW 264.7 cells,and compounds 1,2 and 4 significantly inhibited NO production at 50μmol·L^(-1) CONCLUSION All compounds are isolated from the aerial part of Morinda officinalis How for the first time and compound 1 is a novel one,compounds 1,2 and 4 show potential anti-inflammatory activity.
