摘 要:目的研究萆薢总皂苷(total saponin of Dioscorea,TSD)对高脂血症的调节作用及对动脉粥样硬化(atherosclerosis,AS)的防治作用。方法采用高脂饲料分别建立小鼠、大鼠高脂血症模型,高脂饲料+维生素D_(2)建立大鼠AS模型,测定血脂水平,计算心脏指数、AS指数(AI_(1)、AI_(2))及冠心指数(R-CHR);苏丹Ⅳ染色、HE染色和Masson染色观察大鼠主动脉组织病理学变化。结果与正常组比较,模型组小鼠血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白-胆固醇(LDL-C)显著升高,高密度脂蛋白-胆固醇(HDL-C)显著降低(P<0.01)。TSD高、中剂量组和阿托伐他汀组TC、TG、LDL-C水平明显降低(P<0.05,P<0.01)。在大鼠高脂血症模型观察到类似结果。在大鼠AS模型,模型组血清TC,TG,LDL-C水平升高,HDL-C水平降低,心脏指数、AI_(1)、AI_(2)及R-CHR显著增加(P<0.01),主动脉及主动脉窦部病变明显。TSD高、中剂量组和阿托伐他汀组血清TC,TG,LDL-C含量显著降低,HDL-C含量升高,心脏指数、AI_(1)、AI_(2)及R-CHR显著降低(P<0.05,P<0.01),主动脉及主动脉窦部组织学病变明显改善。结论TSD具有调节血脂和抗AS作用。OBJECTIVE To investigate the regulatory effect of total saponin of Dioscorea(TSD)on hyperlipidemia and its prevention and therapeutic effect of atherosclerosis(AS).METHODS The hyperlipidemia model of mice and rats was established with the high-fat diet,and the rat AS model was established with the high-fat diet plus vitamin D_(2).The blood lipid level was measured,and the cardiac index,AS index(AI_(1),AI_(2))and coronary heart index(R-CHR)were calculated.The pathological changes of aorta were observed by SudanⅣstaining,HE staining and Masson staining respectively.RESULTS Compared with the normal group,the mice in the model group serum total cholesterol(TC),triglyceride(TG)and low density lipoprotein-cholesterol(LDL-C)were significantly increased,while high density lipoprotein-cholesterol(HDL-C)was significantly decreased(P<0.01).The level of TC,TG and LDLC in TSD high and medium dose groups and atorvastatin group were significantly reduced(P<0.05,P<0.01).Similar results were observed in the rat hyperlipidemia model.In the rat AS model,serum TC,TG and LDL-C level in the model group increased,HDLC level decreased,heart index,AI_(1),AI_(2) and R-CHR increased obviously(P<0.01),and aortic and sinus lesions were significant.Serum TC,TG,LDL-C content in TSD high and medium dose groups and atorvastatin group were significantly reduced,HDL-C content was increased,cardiac index,AI_(1),AI_(2) and R-CHR were significantly reduced(P<0.05,P<0.01),and the histological lesions of the aorta and aortic sinus were significantly ameliorated.CONCLUSION TSD has the effects of regulating blood lipid and anti-AS.
