摘　　要：目的:建立高效液相色谱-质谱串联法(LC-MS/MS)测定大活络丸中血竭素的含量。方法:选择Agilent Poroshell 120 EC-C18色谱柱(2.1 mm×100 mm,2.7μm),流动相为甲醇-0.1%甲酸,梯度洗脱,流速0.2 mL/min,柱温为35℃,采用电喷雾离子源(ESI)正离子检测,以多反应监测(MRM)方式扫描。结果:血竭素在37.3~3730.0 ng/mL(r=0.9998)呈良好线性关系;加样回收率(n=6)为100.09%,RSD为1.42%;10批大活络丸中血竭素的含量测定结果为0.29~7.15μg/g。结论:该方法结果准确、简便快速,可用于大活络丸中血竭素的含量测定。Objective:To establish a LC-MS/MS method for the content determination of dracorhodin in dahuoluo pills.Methods:The component was separated on the Agilent Poroshell 120 EC-C18 column(2.1 mm×100 mm,2.7μm)with methanol and 0.1%formic acid as the mobile phase by gradient elution.The flow rate was 0.2 mL/min;The column temperature was set at 35℃,The detection was carried out by electrospray ionization(ESI)in positive modes.Multiple reaction monitoring(MRM)mode was used for quantitative analysis.Results:The linear range was 37.3-3730.0 ng/mL(r=0.9998)for dracorhodin;the average recoveries(n=6)was 100.09%and RSD was 1.42%;the contents in 10 sapmles of dahuoluo pills were 0.29-7.15μg/g for dracorhodin.Conclusion:The method is accurate,rapid and suitable for determination of dracorhodin in dahuoluo pills.
