摘 要:Claudins是构成紧密连接的膜蛋白,参与细胞旁路屏障和通道的形成。在肾脏中,每一段肾小管节段都相应地表达特定的Claudins,这些Claudins赋予该节段独特的细胞旁路离子通透性和选择特性并以此调节水及溶质的转运。此外,一些Claudins还可参与肾脏发生发育。claudins基因突变与多种肾脏疾病有密切关系,如claudin-10b的变异引起HELIX综合征,claudin-16、claudin-19的变异引起家族性低镁血症伴高钙尿症和肾钙质沉着症,claudin-14变异与高钙尿性肾结石高度相关等。本文综述了在肾脏表达的Claudins的结构及其生理功能的研究进展,以及Claudins在健康和肾脏疾病中的作用。Claudins are tight-junction membrane proteins involved in the formation of barriers and pores in the paracellular pathway in epithelial cells.In the kidney,specific Claudins expressed in each segment of renal tubules,which give the segment with unique ion permeability and selectivity to regulate the transport of water and solutes in the paracellular pathway.In addition,some Claudins can also participate in the development of the kidney.Genetic mutations in claudins are related to some kidney diseases.Variants of claudin-10b cause HELIX syndrome,and mutations in claudin-16 and claudin-19 cause familial hypomagnesemia with hypercalciuria and nephrocalcinosis.And mutations in claudin-14 are highly correlated with hypercalciuric kidney stones.This review summarizes the structure and physiological functions of Claudins expressed in the kidney,as well as the role of some Claudins in renal disease.
