摘 要:甘薯双生病毒(sweepoviruses)是侵染甘薯的一类重要病毒,通过烟粉虱以持久方式传播,我国甘薯上至少存在8种甘薯双生病毒。本研究根据我国已报道的8种甘薯双生病毒基因组保守区设计了一组引物,建立了单头烟粉虱中甘薯双生病毒的半巢式PCR快速检测方法。特异性和灵敏性分析结果表明,半巢式PCR具有较高的特异性和灵敏性,以8种不同甘薯双生病毒重组质粒为模板时,该方法能同时检测出8种甘薯双生病毒,且半巢式PCR方法的灵敏性比常规PCR高10~10000倍。对烟粉虱的检测结果表明,该方法能稳定地从单头烟粉虱中检测出甘薯双生病毒。该方法具有经济、快速,灵敏性、特异性和稳定性均较好等优点。Sweepoviruses are the viruses infecting sweet potato and transmitted by Bemisia tabaci in persistent manner.At least eight viruses had been identified in Chinese sweet potato.Primers were designed based on the conserved regions of genome of eight sweepoviruses from China.The rapid semi-nested PCR detection technology was established for the detection of sweepoviruses in single whitefly.The semi-nested PCR had high specificity and sensitivity.With the recombination plasmids of eight sweepoviruses as template,eight viruses could be simultaneously detected.The sensitivity of the semi-nested PCR was higher than that of conventional PCR by 10-10000 times.Sweepoviruses could be steadily detected from individual whitefly by using the semi-nested PCR assay.This assay is of economical,rapid,sensitive,specific and stable for rapid detection of sweepoviruses in B.tabaci.
