摘  要:目的:采用高效液相色谱法测定不同产地、不同生长年限黄芩中6种主要黄酮类化学成分(野黄芩苷、黄芩苷、黄芩素、汉黄芩素、白杨素和千层纸素A)的含量,为全面评价黄芩药材质量提供依据。方法:采集山西运城、山西平顺、河北承德、内蒙古赤峰生长年限分别为1年、2年、3年共12批黄芩样品,黄芩药材粉末经乙醇(每次20 min)超声提取2次,合并提取液。利用岛津C18色谱柱(250 mm×4.6μm,5μm),流动相为乙腈-0.1%甲酸水溶液,梯度洗脱的色谱条件;体积流量1.0 mL·min^(-1),检测波长为274 nm;柱温30℃。结果:所测6种主要有效成分在测定质量浓度范围内线性关系良好,精密度、重复性和稳定性良好;平均回收率为97.85%~99.08%,RSD为0.96%~1.53%。其中野黄芩苷、黄芩苷、白杨素、千层纸素A含量随年限增长而增长,黄芩素、汉黄芩素随年限延长而下降;山西运城、山西平顺、河北承德产黄芩中黄芩苷平均含量远远高于内蒙古赤峰。结论:高效液相色谱法测定黄芩中6种化学成分的含量准确度高,灵敏度好,不同年限和产地黄芩主要有效成分含量不同,为黄芩质量评价提供依据。Objective:To establish a HPLC method to determine the six main chemical constituents in Scutellaria baicalensis(scutellarin,baicalin,baicalein,wogonin,aspen and melaleuca A)from different habitats and different growth years.Methods:Scutellaria baicalensis gathered from Shanxi Yuncheng,Shanxi Pingshun,Hebei Chengde,and Inner Mongolia Chifeng,with 12 groups of samples totally,were simultaneous determination,and the growth years of these Scutellaria baicalensis were 1 year,2 years and 3 years respectively.Powders of Scutellaria baicalensis was extracted twice by ethanol(each for 20 min),and the extracts were merged.Separation was carried out on Ultrasil C18 chromatography co-lumn(250 mm×4.6μm,5μm),by using acetonitrile-0.1%formic acid water as the flow phase for gradient elution;with volume flow at 1.0 mL·min^(-1),detection wavelength at 274 nm and column temperature at 30℃.Results:The linear relation of six main active components measured in the range of mass concentration was good with perfect precision,repeatability and stability.The average recovery rate was 97.85%~99.08%,RSD was 0.96%~1.53%.The contents of scutellarin,baicalin,aspen and melaleuca A increased with the increase of age,while the contents of baicalein and wogonin decreased with the increase of age.The average content of baicalin in Scutellaria baicalensis from Yuncheng in Shanxi,Pingshun in Shanxi and Chengde in Hebei was much higher than that from Chifeng in Inner Mongolia.Conclusion:The HPLC method established for the simultaneous determination of six main chemical compositions in Scutellaria baicalensis is accurate and sensitive;the content of main effective constituents in Scutellaria baicalensis with different habitats and growth years are different.Both of them can provide references for the quality evaluation of Scutellaria baicalensis.
