摘  要:目的:通过水迷宫实验、自主活动实验、听源性惊厥实验和悬尾实验来研究C57BL/6小鼠和C57BL/6 FMR1基因敲除小鼠的行为学差异。方法:通过胚胎复苏获得3只雌性杂合的C57BL/6 FMR1基因敲除小鼠后与C57BL/6小鼠进行繁殖,最终得到纯合的雌性和雄性C57BL/6 FMR1基因敲除小鼠各10只进行行为学实验。结果:在定位航行实验的第四天,KO(基因敲除型)组小鼠的潜伏期明显高于WT(野生型)组(P<0.01);空间搜索实验中,KO组小鼠的超越原平台的次数和平均速度均小于WT组小鼠(P<0.05);在工作记忆实验第四天,KO组小鼠的潜伏期高于WT组小鼠(P<0.05);听源性惊厥实验中,WT组小鼠的惊厥阈值低于KO组小鼠(P<0.01);自主活动实验中,KO组小鼠的自主活动次数高于WT组小鼠(P<0.01);在悬尾实验中,WT组小鼠的静止时间高于KO组小鼠,但不存在显著性差异。结论:在行为学实验中,KO组小鼠受到FMR1基因敲除影响,使KO组小鼠的运动能力和记忆能力明显低于WT组小鼠,更容易在声音的诱导下触发癫痫。Objective:To study the behavioral differences between C57BL/6 mice and C57BL/6 FMR1 knockout mice by Morris water maze,autonomous activity experiment,audiogenic seizures experiment and tail suspension experiment.Methods:Three female heterozygous C57BL/6 FMR1 knockout mice were obtained by embryo resuscitation,and then propagated with C57BL/6 mice.Finally,10 homozygous female and 10 male C57BL/6 FMR1 knockout mice were obtained for behavioral experiments.Results:On the fourth day of positioning navigation experiment,the latency of KO(gene knockout)group was significantly higher than that of WT(wild type)group(P<0.01).In the spatial search experiment,the times and average speed of surpassing the original platform in KO group were lower than those in WT group(P<0.05).In the working memory experiment on the fourth day,the latency of mice in KO group was higher than that in WT group for hours(P<0.05).In audiogenic seizures experiment,the convulsion threshold of WT group mice was lower than that of KO group mice(P<0.01).In the autonomic activity experiment,the number of autonomic activities in KO group was higher than that in WT group(P<0.01).In the tail suspension experiment,the resting time of WT group mice was higher than that of KO group mice,but there was no significant difference.Conclusion:In the behavioral experiment,KO group mice were affected by FMR1 gene knockout,so that the motor ability and memory ability of KO group mice were significantly lower than that of WT group mice,and it was easier to trigger epilepsy under the induction of sound.
