消瘤胶囊对人肝癌耐药细胞株Bel/Fu化疗敏感性的影响

摘　　要：目的:探讨消瘤胶囊对人肝癌耐药细胞株Bel/Fu化疗敏感性的影响。方法:将人肝癌耐药细胞株Bel/Fu随机分为空白对照组、5-氟尿嘧啶组(12.5 mg·L^(-1))、消瘤胶囊组(25 g·L^(-1))、联合用药组(12.5 mg·L^(-1)5-氟尿嘧啶+25 g·L^(-1)消瘤胶囊),每组设置6个复孔。待细胞融合度达80%左右时,各组培养基中加入相应浓度的药物,空白对照组加入等体积细胞培养基,培养48 h。光学显微镜下观察各组细胞状态;MTT法检测细胞增殖抑制率;TUNEL法检测细胞凋亡率;RT-PCR法检测细胞GST-π mRNA、p-gp mRNA水平;Western Blot法检测细胞GST-π、p-gp、gp蛋白表达。结果:与空白对照组比较,5-氟尿嘧啶组、消瘤胶囊组和联合用药组细胞增殖抑制率、细胞凋亡率显著升高(P<0.05,P<0.01),细胞GST-π mRNA、p-gp mRNA水平显著降低(P<0.05),细胞GST-π蛋白表达、p-gp/gp水平显著降低(P<0.05);与5-氟尿嘧啶组比较,消瘤胶囊组和联合用药组细胞增殖抑制率、细胞凋亡率显著升高(P<0.05,P<0.01),细胞GST-π mRNA、p-gp mRNA水平显著降低(P<0.05),细胞GST-π蛋白表达、p-gp/gp水平显著降低(P<0.05);与消瘤胶囊组比较,联合用药组细胞增殖抑制率、细胞凋亡率极显著升高(P<0.01),细胞GST-π mRNA、p-gp mRNA水平显著降低(P<0.05),细胞GST-π蛋白表达、p-gp/gp水平显著降低(P<0.05)。结论:消瘤胶囊可增强Bel/Fu细胞对5-氟尿嘧啶化疗敏感性,抑制细胞增殖、促进细胞凋亡,作用可能与抑制GST-π、p-gp表达有关。Objective:To study the effect of Xiaoliu capsules on chemosensitivity of human drug-resistant hepatic carcinoma Bel/Fu cell line.Methods:The human liver cancer drug-resistant cell line Bel/Fu was randomly divided into a blank control group,5-fluorouracil group(12.5 mg·L^(-1)),Xiaoliu capsule group(25 g·L^(-1)),combined medication group(12.5 mg·L^(-1)5-Fluorouracil+25 g·L^(-1)Xiaoliu Capsule),each group has 6 multiple pores.When the cell fusion reached about 80%,the corresponding concentration of drugs was added to the culture medium of each group,and a blank control group was added an equal volume of common cell culture medium,incubated for 48 h.The cell status of each group was observed under an optical microscope;MTT method was used to detect cell proliferation inhibition rate;TUNEL method was used to detect cell apoptosis rate;RT-PCR method was used to detect cell GST-π mRNA and p-gp mRNA levels;Western Blot method was used to detect cell GST-π,p-gp,gp protein expression.Results:Compared with the blank control group,the cell proliferation inhibition rate and apoptosis rate of 5-fluorouracil group,Xiaoliu capsule group and combination medication group were significantly increased(P<0.05,P<0.01),and the levels of cellular GST-πmRNA and p-gp mRNA Significantly reduced(P<0.05),cell GST-πprotein expression and p-gp/gp levels were significantly reduced(P<0.05);Compared with the 5-fluorouracil group,the cell proliferation inhibition rate and apoptosis rate of the Xiaoliu capsule group and the combination medication group were significantly increased(P<0.05,P<0.01),and the levels of cellular GST-πmRNA and p-gp mRNA were significantly reduced(P<0.05),cell GST-πprotein expression and p-gp/gp levels were significantly reduced(P<0.05);Compared with the Xiaoliu Capsule group,the cell proliferation inhibition rate and apoptosis rate of the combined drug group were significantly increased(P<0.01),the levels of cellular GST-πmRNA and p-gp mRNA were significantly reduced(P<0.05),and the cell GST-πProtein expression
