摘  要:地被菊(Chrysanthemum morifolium)是菊科菊属多年生宿根草本植物,花型整齐,色彩多样,有较高的观赏价值。北方地区温度低,严重限制了地被菊的栽培范围。碱性螺旋-环-螺旋(basic helix-loophelix, bHLH)转录因子家族广泛参与植物的生长发育和非生物胁迫响应。本研究以地被菊’映洁’为材料,根据转录组数据克隆了17个CmbHLHs基因,ORF在603~1 242 bp之间,对蛋白理化性质进行预测分析发现,17个CmbHLHs蛋白中8个蛋白为酸性,9个蛋白为碱性,这些蛋白都定位在细胞核上。bHLH蛋白二级结构α-螺旋占比为25.49%~51.50%,β-转角占比为1.42%~5.88%;miRNA预测分析显示,9个miRNA可能调控8个CmbHLHs基因转录后的表达。利用MEME软件预测CmbHLH基序组成,共鉴定出6个保守基序,除CmbHLH8和CmbHLH15外,其他蛋白都包含基序1和基序2。系统发育分析显示,17个CmbHLHs基因分布于12个bHLH亚家族。低温处理地被菊幼苗,qPCR分析发现,7个CmbHLHs基因受到低温胁迫诱导后表达量增加。本研究可为地被菊bHLH基因功能的进一步研究提供参考。Ground cover chrysanthemum(Chrysanthemum morifolium) is a perennial herb of the genus Chrysanthemum in the Compositae family, with neat flowers and diverse colors, which has high ornamental value. The low temperature in the north severely limits the cultivation range of ground cover chrysanthemum.The basic helix-loop-helix(bHLH) transcription factors are widely involved in plant growth and development and abiotic stress response. In this study, ground cover chrysanthemum ’Yingjie’ was used as material, and 17 CmbHLHs were cloned based on transcriptome data, with ORF between 603 bp and 1 242 bp, and their physical and chemical properties were predicted. The physical and chemical properties of 17 CmbHLHs showed that 8 proteins were acidic and 9 proteins were basic, and these proteins were all located on the nucleus;The proportion of α-helix in the secondary structure of bHLH protein was 25.49%~51.50%, and the proportion of β-turn was 1.42%~5.88%;miRNA prediction analysis showed that 9 miRNAs might regulate the post-transcriptional expression of 8 CmbHLHs. Using MEME to predict the composition of CmbHLH motifs,a total of 6 conserved sequences were identified, except CmbHLH8 and CmbHLH15, all the other proteins contained motif 1 and motif 2. Phylogenetic analysis showed that 17 CmbHLHs were distributed in 12 bHLH subfamilies. Ground cover chrysanthemum seedlings were treated with low temperature, qPCR analysis found that the expression of 7 CmbHLHs increased under low temperature stress. This study provides references for further research on the function of ground cover chrysanthemum bHLH gene.
