摘  要:草原龙胆(Eustoma grandiflorum)花色典雅、花型丰富,是全球最受欢迎的切花之一。本研究以草原龙胆‘阿琳娜’叶片为外植体,研究了6-BA、NAA、GA_(3)和IBA不同组合对其出芽、壮苗、增殖、生根的影响。结果表明:最佳不定芽诱导培养基为MS+6-BA 0.5 mg/L+NAA 0.05 mg/L,不定芽诱导率达90.0%,平均出芽数为4.52个;MS培养基可实现壮苗;最佳增殖培养基为MS+6-BA 0.2 mg/L+IBA 0.2 mg/L+GA_(3)0.3 mg/L,增殖系数为6.41;培养基1/2MS+IBA 0.5 mg/L有利于生根。初步建立了草原龙胆‘阿琳娜’高效再生体系,为草原龙胆优良品种快速繁殖及基因工程研究提供了依据。Lisianthus(Eustoma grandiflorum)is one of the most popular cut flowers in the world,with elegant colors and rich flower patterns.In this study,leaves of lisianthus’Arena’were used as explants to study the effects of different combinations of 6-BA,NAA,GA_(3)and IBA on budding,strong seedling,proliferation and rooting.Results showed that the optimum adventitious bud induction medium was MS+6-BA 0.5 mg/L+NAA 0.05 mg/L,the adventitious bud induction rate was 90.0%,and the average number of buds was 4.52.The MS medium could be used for strong seedling.The optimum proliferation medium was MS+6-BA 0.2 mg/L+IBA 0.2 mg/L+GA_(3)0.3 mg/L,and the proliferation coefficient was 6.41.The medium with 1/2MS+IBA 0.5 mg/L was good for rooting.Thus,a high-efficiency regeneration system of lisianthus’Arena’was preliminarily established,which provided a basis for the rapid propagation and genetic engineering breeding of elite varieties of lisianthus.
