摘 要:目的研究黄药子醇提物对白血病CEM细胞生长抑制及其可能机制。方法生长状态良好的CEM细胞分组培养:对照组,4个不同浓度黄药子醇提物组。CCK8法检测细胞增殖抑制率;倒置显微镜,油镜下观察细胞形态及结构的改变;DNA琼脂凝胶电泳观察细胞凋亡;Western blot(WB)法检测Caspase3、Caspase8的表达水平。结果黄药子醇提物能显著抑制CEM细胞的增殖,相较于对照组,黄药子组对细胞的抑制作用明显,并呈时间、剂量相关性,黄药子醇提物作用CEM细胞24、48、72h后的IC50分别为214.6、125.1、96.43μg/ml;倒置显微镜、油镜下可见各药物组细胞体积明显缩小,细胞膜皱缩,细胞核出现程度不等的裂解;细胞凋亡检测结果发现,各浓度的黄药子组均产生细胞凋亡即产生DNA条带;WB结果表明黄药子醇提物可以使Caspase3和Caspase8蛋白表达水平升高。结论黄药子醇提物可以明显抑制CEM细胞的增殖同时诱导其凋亡,其机制可能与上调Caspase3和Caspase8蛋白的表达有关。Objective To study the effect of ethanol extract of Dioscorea bulbifera Linn. on the proliferation of leukemia CEM cells and its possible mechanism. Methods CEM cells were cultured in vitro and grouped into groups, including control group and ethanol extract of Dioscorea bulbifera Linn. with different concentrations. CCK8 method was used to detect the cell proliferation inhibition rate;the influence of the ethanol extract of Dioscorea bulbifera Linn. on the morphology of CEM cells was observed under inverted microscope and oil microscope;the apoptosis of CEM cells after the ethanol extract of Dioscorea bulbifera Linn. was treated by DNA gel electrophoresis was observed;Western Blot(WB) method was used to detect the expression of related apoptosis proteins Caspase3 and Caspase8 after the ethanol extract of Dioscorea bulbifera Linn. acted on CEM cells.Results The ethanol extract of Dioscorea bulbifera Linn can significantly inhibit the proliferation of CEM cells. Compared with the control group, the ethanol extract of Dioscorea bulbifera Linn. group has a significant inhibitory effect on the cells, and was related to time and dose. The IC50 of each drug group was 214.6, 125.1, 96.43μg/ml respectively;under the inverted microscope and oil microscope, it was seen that the cell volume of each drug group was significantly reduced, the cell membrane was shrunk, and the nucleus appeared to be lysed to varying degrees;the apoptosis test results found that the concentration of the ethanol extract of Dioscorea bulbifera Linn. can increase the protein expression level of Caspase3 and Caspase8.Conclusion The ethanol extract of Dioscorea bulbifera Linn. can significantly inhibit the proliferation of CEM cells and induce their apoptosis. The mechanism may be related to the up-regulation of the expression of Caspase3 and Caspase8 proteins.
