摘  要:目的旨在使用网络药理学的方法探讨柴贝止痫汤入血成分治疗癫痫的潜在机制。方法通过检索相关数据库收集治疗癫痫的靶点以及柴贝止痫汤已鉴定的15个入血成分的作用靶点,构建柴贝止痫汤入血成分治疗癫痫的靶点网络图,并检索靶点间的蛋白-蛋白相互作用。使用Cytoscape构建网络图、分析网络。并将共有靶点使用clusterProfiler包进行GO富集分析和KEGG富集分析。结果柴贝止痫汤中的入血成分具有125个治疗癫痫的靶点,原儿茶醛具有较多的癫痫治疗靶点,碳酸酐酶是柴贝止痫汤入血成分的主要治疗靶点,AKT1是治疗癫痫的关键靶点。治疗靶点富集于3744个生物过程,295个细胞组分,431个分子功能和253个KEGG通路。主要作用于神经元突触和神经元胞体,在氧化应激、神经细胞死亡、神经递质反应等中发挥治疗作用,并与cAMP信号通路有关。结论柴贝止痫汤可能是通过调节神经递质反应、氧化应激、神经细胞死亡等发挥治疗癫痫的作用。仍需进一步的实验研究验证该结果。Objective In this study, we deciphered the pharmacology of components in Chaibei Zhixian Decoction absorbed into blood(CCZD) for epilepsy based on network pharmacology.Methods By searching the relevant database we collected the targets of epilepsy and the targets of 15 components absorbed into blood identified in Chaibei Zhixian decoction. The target network of CCZD for epilepsy was constructed, and the protein-protein interaction between the targets were searched. Cytoscape software was used to build the network and analyze the network. The clusterProfiler package was used for GO enrichment analysis and KEGG enrichment analysis. Results In CCZD There are 125 therapeutic targets for epilepsy, 3,4-Dihydroxybenzaldehyde had the most therapeutic targets for epilepsy. Carbonic anhydrase is the main therapeutic target for epilepsy and AKT1 is the key target for epilepsy. The therapeutic target was enriched in 3744 biological processes, 295 cell components, 431 molecular functions and 253 KEGG pathways. It plays a therapeutic role in oxidative stress, neuronal cell death, neurotransmitter response, and is related to cAMP signaling pathway. Conclusion CCZD may play a role in the treatment of epilepsy by regulating neurotransmitter response, oxidative stress, and nerve cell death. Further experimental studies are needed to verify the results.
