摘  要:目的探讨电针刺激足阳明胃经穴位及阿是穴对脊髓损伤大鼠神经功能恢复的影响及受损脊髓中c-fos、c-jun的mRNA、蛋白表达变化。方法运用钳夹法建立脊髓损伤(SCI)模型;120只模型大鼠随机分为3组:模型对照组,阳明经组(电针刺激足阳明胃经穴位),阿是穴组(电针刺激阿是穴);每组依据留取脊髓样本时间点分为1、2、3、4、5周亚组。对各组动物进行BBB运动功能评分,qPCR、免疫组化以及Western blot检测各组大鼠受损脊髓中c-fos/c-jun mRNA及蛋白的表达。结果阳明经组和阿是穴组的BBB评分显著高于模型对照组(P<0.05);阳明经组和阿是穴组c-fos及c-jun的mRNA及蛋白表达在前2周均显著低于模型对照组(P<0.05);在3~4周阳明经组c-fos及c-jun的mRNA及蛋白表达较模型对照组显著降低(P<0.05);第5周时三组无显著差异(P>0.05)。结论电针刺激可能通过下调SCI大鼠受损脊髓节段c-fos及c-jun的mRNA及蛋白表达促进其神经功能的恢复。Objective To investigate the effect of electrical acupuncture at the acupoints of the Stomach Meridian of Foot Yangming and Ashi point on the recovery of nerve function in rats with spinal cord injury and the expression changes of c-fos and c-jun mRNA and protein in the injured spinal cord.Methods The spinal cord injury(SCI)model was established by clamp method;120 model rats were randomly divided into 3 groups:model control group,Yangming meridian group(electrical acupuncture to stimulate the acupoints of the foot Yangming stomach meridian),Ashi point group(Electrical acupuncture to stimulate Ashi point);each group was divided into subgroups of 1,2,3,4,and 5 weeks according to the time point of the spinal cord sample obtained.BBB motor function scores were performed on each group of animals,and the expression of c-fos/c-jun mRNA and protein in the injured spinal cord of each group of rats was detected by qPCR,immunohistochemistry and Western blot.Results Compared with the control group,the BBB scores of the Yangmingjing group and Ashi point group were significantly high(P<0.05);the mRNA and protein expression of c-fos and c-jun in the Yangmingjing group and Ashi point group were significantly high compared the model control group in the first two weeks(P<0.05).The expressions of mRNA and protein of c-fos and c-jun in the Yangming Jing group of 3-4 weeks were significantly lower than those of the model control group(P<0.05).At the fifth week,there was no significant difference among the three groups(P>0.05).Conclusion Electroacupuncture can down regulate the mRNA and protein expression of c-fos and c-jun in injured spinal cord segments of SCI rats,and promote the recovery of neurological function in SCI rats.
