摘 要:环状RNA(circRNA)是一类呈环状结构的内源性非编码RNA。通过对3只雄性梅花鹿鹿茸间充质组织的小鞍子时期和快速生长期进行高通量测序,根据测序结果鉴定其中的环状RNA,分析不同发育阶段环状RNA表达量差异,对差异表达的环状RNA来源基因进行GO富集分析和KEGG功能注释分类,最终构建了与鹿茸生长发育相关差异最显著的circRNA-miRNA-mRNA调控网络。结果表明:鉴定得到7790个环状RNA,其中,从鹿茸发育前期到鹿茸的快速生长期鉴定得到208个显著差异表达的环状RNA,包括39个显著上调差异表达的环状RNA以及169个显著下调差异表达的环状RNA。环状RNA显著富集到生物调控、细胞过程、代谢过程、细胞、催化活性等多种GO条目,显著富集到粘着斑、PI3K-Akt信号通路、MAPK信号通路、TNF信号通路、FoxO信号通路。最终筛选的最显著表达的10个环状RNA构建了与鹿茸发育相关的circRNA-miRNA-mRNA互作调控网络。Circular RNA(circRNA)is an endogenous non-coding RNA with a circular structure.Herein,when antlers grew to a saddle-like appearance and grew with two branches,the mesenchymal tissue of three male sika deer antlers were selected for high-throughput sequencing.circRNA was identified based on the high-throughput sequencing,the expression differenceswas analyzed in circRNA at different periods,and GO and KEGG enrichment analysis was carried out.Finally,for exploring the role of circRNA in the antler growth,the circRNA-miRNA-mRNA regulatory network related to antler growth with the most significant difference was constructed.A total of 7790 circRNA were identified and 208 significantly differentially expressed circRNA were identified,including 39 up-regulated differentially expressed circRNA and 169 down-regulated differentially expressed circRNA.GO enrichment analysis significantly enriched in various GO items such as biological regulation,cellular processes,metabolic processes,cells,and catalytic activity,and the differentially expressed circRNAsignificantly enriched in focal adhesions,PI3K-Akt signaling pathway,MAPK signaling pathway,TNF signaling pathway and FoxO signaling pathway.Finally,the 10 most differentially expressedcircRNAs were screened to construct a circRNA-miRNA-mRNA interaction regulatory network related to antler growth.
