半枝莲种子质量检验方法的研究

摘　　要：目的研究半枝莲种子的质量检验方法,为制定半枝莲种子质量检验规程和分级标准提供依据。方法参照《农作物种子检验规程(GB/T 3543-1995)》,通过测定扦样、净度、真实性、千粒重、含水量、种子萌发条件以及生活力等指标研究半枝莲种子质量检验的相关方法。结果半枝莲种子扦样送检样品最少为8.0 g,净度分析式样最少为0.8 g;可依据外观形态判断半枝莲种子真实性;种子质量测定采用千粒法;种子含水量测定采用整粒种子高温恒温烘干法:烘干温度为(131±1)℃,烘干时长为0.5 h;种子萌发时发芽床选用双层纱布,光照/黑暗各12 h交替条件下25℃恒温培养14 d;采用电导率法测定种子生活力。结论建立的方法操作简便、重复性好,适用于半枝莲种子的质量检验。Objective To provide a basis for establishing seed testing rules and seed quality standard of Scutellaria barbata D.Don.Methods The testing methods of sampling,authenticity and purity,thousand seed weight,moisture,germination rate and viability for seed quality of S.barbata are studied according to the Rules for Agricultural Seed Testing(GB/T 3543-1995).Results The sampled seed of S.barbata for testing is at least 8.0 g,and the sample for purity analysis is at least 0.8 g.The verification of S.barbata seeds could be authenticated by phenotype.The weight of the seed is estimated by 1000-seed method.The moisture of S.barbata seeds is determined by higher temperature method:the drying temperature is(131±1)℃and the drying time is 0.5 h.The germination methods of S.barbata seeds are also optimized.The germination temperature is set at 25℃and the bilayers gauze is chosen as germinating bed.The light and the darkness alternated every 12 h.After 14 d,the germination rate of S.barbata seeds could be calculated.Compared with TTC method and BTB method,the electrical conductivity measurement is more suitable for testing the viability of S.barbata seeds.Conclusion The established method is reproducible and easy to operate,which is suitable for the quality inspection of S.barbata seeds.
