摘　　要：目的建立由长期饮酒导致的简便、稳定、实用的高血压伴肝损伤病变小鼠模型。方法100只SPF级昆明种小鼠经标准饲料适应性喂养1 w后随机均分为模型组和对照组。模型组采用自由饮酒法,普通饲料喂养72 d;对照组自由饮水,普通饲料喂养72 d。实验中测定食量、体重、自主活动度、抓力、尾动脉收缩压、舒张压、平均动脉压。同时电镜下观察肝组织病变情况并测定肝功能4项指标。结果模型组血压显著升高,电镜下可见肝细胞水肿、脂肪变性伴坏死等病理特征,且谷丙转氨酶(ALT)、谷草转氨酶(AST)等指标显著高于对照组(P<0.01)。结论以自由饮酒法搭配普通饲料喂养是一种简单快速的高血压合并肝损伤小鼠模型的建立方法。Objective To establish a simple,stable and practical mouse model of hypertension complicated with liver injury caused by long-term alcohol consumption.Methods A total of 100 SPF Kunming mice were randomly divided into model group and control group after adaptive feeding with standard diet for 1 week.The model group mice were allowed to drink alcohol freely and raised with common feed for 72 days.The control group mice were allowed to drink water freely and raised with common feed for 72 days.Food intake,body weight,spontaneous locomotor activity,grip strength,tail systolic blood pressure,diastolic blood pressure and mean arterial pressure were measured.At the same time,the pathological changes of liver tissue were observed under electron microscope and 4 indexes of liver function were determined.Results The blood pressure of model group was significantly increased,and the pathological features of hepatic cell edema,steatosis and necrosis were observed under electron microscope.Compared with control group,ALT,AST and other indexes in model group were significantly increased.Conclusions Free access to alcohol combined with common feed is a simple and rapid method for establishing a mouse model of hypertension combined with liver injury.
