摘  要:目的探讨杜仲黄酮对2型糖尿病小鼠胰腺凋亡因子的影响。方法通过链脲佐菌素(STZ)和高脂高糖饮食诱导构建糖尿病小鼠模型。将糖尿病模型小鼠随机分为模型组、二甲双胍组、杜仲黄酮低、高剂量组。另以健康昆明小鼠为空白组。各组予相对应药物灌胃60 d,每日1次。空白对照组和模型组予等体积生理盐水灌胃。末次给药后,小鼠禁食不禁水12 h,测其空腹血糖(FBG)。取部分胰腺组织进行TUNEL染色,观察其细胞凋亡情况。ELISA法检测血清中空腹C-肽的含量以及胰腺中氧自由基(ROS)、炎症因子白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、跨膜蛋白Fas、跨膜蛋白配体Fasl的含量。Western blot检测胰腺中促进细胞凋亡因子Bax和抑制细胞凋亡因子Bcl-2表达。结果与模型组比较,杜仲黄酮组小鼠空腹血糖下降(P<0.05);胰腺组织病理形态改善,血清中C-肽含量升高(P<0.05);胰腺中ROS、IL-6、TNF-α、Fas、Fasl含量及Bax表达减少,Bcl-2表达增加(P<0.05)。结论杜仲黄酮能降低糖尿病小鼠血糖及抑制胰腺细胞凋亡,其作用机制可能与调节胰腺组织中相关凋亡因子表达,减少炎症因子含量有关。Objective To investigate the effect of flavones from eucommia bark on pancreatic apoptosis factors in type 2 diabetic mice.Methods A diabetic mouse model was established by streptozotocin and the high-fat and high-sugar diet induction.The diabetic model mice were then randomly divided into the model group,the metformin group,the low and high-dose groups of eucommia bark flavones.In addition,healthy Kunming mice were used as the blank group.Each group was given the corresponding drugs by gavage for 60 days,once a day.The blank control group and the model group were given an equal volume of normal saline by gavage.After the last administration,the mice were fasted for 12 h,and their fasting blood glucose (FBG) was measured.Some pancreatic tissues were stained with TUNEL to observe the apoptosis.The contents of fasting C-peptide in serum,oxygen free radical (ROS),inflammatory factor interleukin-6 (IL-6),tumor necrosis factor-α (TNF-α),transmembrane protein Fas and transmembrane protein ligand Fasl in pancreas were detected by ELISA.The expression of Bax and Bcl-2 was detected by Western blot.Results Compared with the model group,the fasting blood glucose of the mice in the eucommia bark flavone group was decreased (P<0.05).The pathological morphology of pancreatic tissues was improved,and the serum C-peptide content was increased (P<0.05).The contents of ROS,IL-6,TNF-α,Fas,Fasl and the expression of Bax in pancreas were decreased,while the expression of Bcl-2 was increased (P<0.05).Conclusion Flavones extracted from eucommia bark can reduce the blood glucose and inhibit the apoptosis of pancreatic cells in diabetic mice,whose mechanism may be related to regulating the expression of apoptosis factors and reducing the content of inflammatory factors in pancreatic tissues.
