摘 要:目的研究蒙药复方舒心康(SXK)对异丙肾上腺素(ISO)引起的大鼠急性心肌缺血的治疗效果。方法将SD大鼠随机分为空白组、模型组、阳性组(地奥心血康42 mg/kg)、SXK高(40.0 mg/kg)、中(20.0 mg/kg)、低剂量组(10.0 mg/kg),每组10只。阳性组及SXK各剂量组灌胃给药,空白组和模型组给予等量蒸馏水,连续15 d。从第13天开始,除了空白组,其他各组按5 mg/kg剂量腹腔注射异丙肾上腺素,每日1次,连续3 d,观察心电图(ECG)J点位移及T波高度变化,检测大鼠血清中谷胱甘肽过氧化物酶(GSH-Px)的活性、超氧化物歧化酶(SOD)含量和丙二醛(MDA)含量,观察大鼠心肌组织病理形态学的变化。结果与模型组相比,SXK高剂量能显著改善J点和T波变化幅度(P<0.05);SXK高、中、低3个剂量组大鼠血清中GSH-Px、SOD活力均有一定程度升高,MDA 含量降低,SXK高剂量组大鼠血清中GSH-Px活力升高(P<0.05),大鼠血清中MDA 含量下降(P<0.05),同时,能明显减轻病理心肌的损伤程度。结论 SXK能有效抑制异丙肾上腺素所致大鼠心肌组织缺血,保护心肌细胞免受损伤。Objective To study the therapeutic effect of the Mongolian medicine compound Shuxinkang (SXK) on acute myocardial ischemia in rats induced by isoprenaline (ISO).Methods The healthy SD rats were randomly divided into a blank group,a model group,a positive group (Di’ao Xinxuekang 42 mg/kg),SXK high-dose (40.0 mg/kg),medium-dose (20.0 mg/kg) and low-dose (10.0 mg/kg) groups,with 10 rats in each group.The positive group and the SXK groups were given the corresponding drugs by gavage respectively,and the blank group and model group were given the same amount of distilled water,all for 15 consecutive days.From the 13th day on,all the groups except the blank group were intraperitoneally injected with isoprenaline at a dose of 5 mg/kg,once a day,for 3 consecutive days.The changes of J-point displacement and T-wave height of electrocardiogram (ECG) were observed,and the activity of glutathione peroxidase (GSH-Px),the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in serum of rats were detected,and the pathological changes of the rat myocardial tissues were observed.Results Compared with the model group,the high dose of SXK significantly improved the change amplitude of the J point and T wave (P<0.05).The activity of GSH-Px and SOD in serum of the rats in the three SXK groups was increased to a certain extent,while the content of MDA was decreased,among which the activity of GSH-Px in serum of the rats in the SXK high-dose group was increased significantly (P<0.05),while the content of MDA in serum was decreased significantly (P<0.05).At the same time,the high dose of SXK significantly reduced the injury degree of pathological myocardium.Conclusion SXK can effectively inhibit the Iso-induced myocardial ischemia in rats,protect myocardial cells from damage,thus playing a role in the treatment of ischemic heart disease.
