摘  要:目的系统评价花生四烯酸-5-脂加氧酶(ALOX5)基因启动子SP-1结合位点基因多态性与支气管哮喘的相关性。方法计算机检索PubMed、Embase、知网、万方、维普、中国生物医学文献等数据库,纳入有关ALOX5基因启动子SP-1结合位点基因多态性与哮喘相关性的病例对照研究,提取相关数据并进行文献质量评价后,采用Review Manager 5.3软件进行Meta分析。结果共纳入6篇文献进行研究,包括哮喘病例722例(病例组)与非哮喘病例626例(对照组)。Meta分析结果显示,病例组ALOX5基因启动子SP-1结合位点6R等位基因的分布频率>对照组,5R等位基因的分布频率<对照组(均P<0.05),而病例组和对照组4R及3R等位基因的分布频率均无统计学差异与哮喘发病无相关性(均P>0.05)。结论 ALOX5基因启动子SP-1结合位点基因多态性(6R等位基因和5R等位基因)与哮喘发病相关,今后需进一步探讨其在哮喘易感人群筛查和哮喘患者的临床分型及药物基因组学研究中的作用。Objective To systematically evaluate the correlation between arachidonate 5-lipoxygenase(ALOX5) gene promoter SP-1 binding site gene polymorphism and bronchial asthma.Methods A computer-based retrieval was performed in PubMed, Embase, CNKI, Wanfang Data, VIP, CBM and other databases to enroll the case-control studies on the correlation between ALOX5 gene promoter SP-1 binding site gene polymorphism and asthma. After the related data extraction and literature quality assessment, Review Manager 5.3 software was employed to perform a Meta-analysis. Results A total of six articles were included for the study, including 722 asthma cases(case group) and 626 non-asthma cases(control group). The results of Meta-analysis revealed that the case group exhibited an increased distribution frequency of 6 R allele at ALOX5 gene promoter SP-1 binding site and a reduced distribution frequency of 5R allele as compared with the control group(all P<0.05);whereas the distribution frequency of 4R or 3R allele showed no statistically significant difference between the case group and the control group and did not correlate with asthma attack(all P>0.05). Conclusion ALOX5 gene promoter SP-1 binding site gene polymorphism(6R allele and 5R allele) correlates with asthma attack. Its role in the screening of population who are susceptible to asthma as well as in asthmatic patients′ clinical classification and pharmacogenomic research should be investigated further in future.
