摘 要:目的探讨与分析人尿激酶原酶对心肌梗死大鼠血清淀粉样蛋白A(SAA)和脂蛋白相关磷脂酶A2(LP-PLA2)的影响。方法48只清洁级SD雄性大鼠购自北京维通利华公司,体重230g左右,均给予建立心肌梗死模型。其中42只心肌梗死大鼠按照随机数字表法分为3组-模型组、实验1组、实验2组,每组各14只。实验1组、实验2组分别给予灌胃人尿激肽原酶1、2mg/kg,模型组给予等量0.9%氯化钠溶液灌胃,1次/d,共4周。测定左心室舒张末期容积(LVEDV)、收缩末期容积(LVESV)、等容收缩期左心室内压力上升最大速率(dP/dtMax)与最小速率(dP/dtMin);计算心脏重量与大鼠体重比值(HW/BW)和心尖部心肌组织厚度;采用酶联免疫吸附试验法检测血清SAA和LPPLA2含量。结果实验1组、实验2组治疗第2周与第4周的LVEDV、LVESV均低于模型组,dP/dtMax与dP/dtMin值高于模型组,差异均有统计学意义(P<0.05),实验1组与实验2组对比差异无统计学意义(P>0.05)。实验1组、实验2组治疗第4周的心肌厚度与HW/BW比值均高于模型组,心脏血清SAA和LP-PLA2含量低于模型组,差异均有统计学意义(P<0.05),实验1组与实验2组对比差异无统计学意义(P>0.05)。结论人尿激酶原酶在心肌梗死大鼠的应用能抑制血清SAA和LP-PLA2的释放,改善心功能与血流动力学状况,从而提高大鼠的心肌厚度、HW/BW比值。Objective To explore and analysis the effects of human urinary kallidinogenase on serum amyliod A(SAA)and lipoprotein-associated phospholipase A2(LP-PLA2)in rats with myocardial infarction.Methods Forty-eight clean SD male rats were purchased from Beijing Weitong Lihua Company,weighing about 230 g,and all were given myocardial infarction models.Forty-two myocardial infarction rats were divided into 3 groups according to the random number table method-model group,experiment 1 group,and experiment 2,with 14 rats in each group.The experimental group 1 and experimental group 2 were given human urinary kininogenase 1 mg/kg and 2 mg/kg by gavage,and the model group were given the same amount of 0.9%normal saline by gavage,once daily for 4 weeks.Left ventricular end-diastolic volume(LVEDV),left ventricular end-systolic volume(LVESV),the maximum rate(dP/dtMax)and minimum rate(dP/dtMin)of left ventricular pressure rise during isovolumic systole were measured;the ratio of heart weight to rat body weight(HW/BW)and apical myocardial tissue thickness were calculated;serum SAA and LP-PLA2 levels were detected by enzyme-linked immunosorbent assay.Results The LVEDV and LVESV in the 2nd and 4th weeks of treatment in the experimental group 1 and experimental group 2 were lower than those in the model group,dP/dtMax and dP/dtMin values were higher than those in the model group,the differences were statistically significant(P<0.05),there were no significant differences compared between the experimental group 1 and experimental group 2(P>0.05).The myocardial thickness and HW/BW ratio of the experimental group 1 and experimental group 2 at the 4th week of treatment were higher than those of the model group,and the serum SAA and LP-PLA2 contents of the heart were lower than those of the model group,the differences were statistically significant(P<0.05),there were no significant differences compared between the experimental group 1 and experimental group 2(P>0.05).Conclusion The application of human urinary kallidinogenase in rats with
