基于网络药理学方法探究三七接骨丸治疗骨质疏松症的作用机制

摘　　要：目的:探究三七接骨丸治疗骨质疏松症的潜在靶点及其作用机制。方法:应用数据库中医药整合药理学网络计算研究平台筛选出本处方5味中药潜在活性成分及作用靶点;从GeneCards、TTD和OMIM数据库查询骨质疏松症疾病靶点;分别采用韦恩图取得交集,运用Cytoscape 3.8.0软件构建"药物-化合物-靶点"作用网络,运用STRING软件构建蛋白质-蛋白质相互作用网络,对所得蛋白质-蛋白质相互作用网络进行拓扑分析,筛选发挥治疗作用的关键靶点;同时利用DAVID数据库、KEGG分析平台进行GO分析和KEGG信号通路分析。结果:①文章得到三七接骨丸治疗骨质疏松症的有效成分34个、靶点114个,关键成分有槲皮素、山柰酚及豆甾醇等;②GO和KEGG分析结果显示,三七接骨丸可能通过影响P13K-Akt信号通路、HIF-1信号通路、肿瘤坏死因子信号通路和白细胞介素17信号通路等主要通路,集中于新陈代谢、细胞增殖、细胞存活、生长和血管生成等功能来发挥防治骨质疏松症的作用;③结果证实三七接骨丸通过多成分-多靶点-多途径的治疗特点干预骨质疏松症。结论:本研究说明了三七接骨丸治疗骨质疏松症的作用机制,为三七接骨丸进一步的基础研究以及临床应用提供了新的思路与线索。Objective:To explore the potential targets of Sanqi Jiegu Wan(三七接骨丸)in treating osteoporosis and its action mechanism.Methods:The potential active ingredients and targets of 5 traditional Chinese medicines in this prescription were screened in the integrated pharmacology of traditional Chinese medicine network computational research platform.The osteoporosis disease targets were inquired in GeneCards,TTD and OMIM databases,and their intersection was obtained respectively by Venn diagrams.A‘medicine-compound-target’interaction network was construct by Cytoscape 3.8.0 software,and a PPI network was established by STRING software.At the same time,a GO analysis and a KEGG signal pathway analysis were executed by DAVID database,KEGG analysis platform respectively.Results:In the included literature,34 active ingredients and 114 targets of treating osteoporosis with Sanqi Jiegu Wan were obtained.The key ingredients are quercetin,kaempferol and stigmasterol,etc..The GO and KEGG analysis results showed that Sanqi Jiegu Wan may play a role in preventing and treating osteoporosis by affecting the main pathways such as P13 KAkt signaling pathway,HIF-1 signaling pathway,tumor necrosis factor signaling pathway,and interleukin 17 signaling pathway,and focusing on metabolism,cell proliferation,cell survival,cell growth,angiogenesis and other functions.The results confirmed that Sanqi Jiegu Wan treated osteoporosis through multi-component,multi-target,and multi-channel intervention.Conclusion:This study illustrates the mechanism of Sanqi Jiegu Wan in treating osteoporosis,and provides new ideas and clues for the further basic research and clinical application of Sanqi Jiegu Wan.
