摘 要:目的:采用星点设计-效应面法优化宣肺解毒胶囊水提取工艺。方法:以料液比、两次提取时间为考察因素,以连翘苷提取率及出膏率“归一化”后总评OD值为考察指标,采用Design-Expert 8.0.6.1软件设计试验,应用多元线性方程建立考察因素与效应面之间的数学模型,绘制考察因素交互影响的等高线图和效应面图,优化宣肺解毒胶囊中最佳提取工艺并进行验证。结果:宣肺解毒胶囊水提取中药组分最佳工艺条件为料液比9 mL/g,第一次提取时间170 min,第二次提取时间80 min,所得预测值总评OD值1.16,经验证后实际得总评OD值1.124,与预测值偏差-3.6%,所建立的数学模型预测性良好。结论:星点设计-效应面法所优化的宣肺解毒胶囊水提取中药组分最佳工艺条件稳定,考察指标在可控范围最大,实际操作中具有一定可行性。Objective:To optimize the water extraction process of Xuanfei Jiedu capsule(宣肺解毒胶囊)by Central Composite Design-Response Surface Method.Methods:The liquid ratio of material and the extraction time were taken as the observation factors,the OD value of the extraction rate of forsythiol glycosides and the paste yield after"normalization"was taken as the investigation index.Using design-expert 8.0.6.1 software Design experiment,the mathematical model between the factors and the effect surface was established by using multiple linear equations,the contour map and effect surface map of the interaction between the factors were drawn,and the optimal extraction process of Xuanfei Jiedu capsule was optimized and verified.Results:The optimal extraction conditions were as follows:the ratio of solid to liquid was 9 mL/g,the first extraction time was 170 min,and the second extraction time was 80 min.The predicted OD value was 1.16.After verification,the actual OD value was 1.124,which was-3.6%different from the predicted value.Conclusion:The optimal process condition of Xuanfei Jiedu capsule water extraction is stable,the index is the largest in the controllable range,and it is feasible in practice.
