摘  要:目的探究孕期和哺乳期暴露重铬酸钾对仔鼠卵巢损伤的影响。方法2019年7月将体重、毛色、生理状态相似的7周龄无特定病原体级SD大鼠雌鼠15只分为4组,分别为低(1/8 LD50)、中(1/4 LD50)、高(1/2 LD50)浓度染毒组(各4只)和对照组(3只)。对照组给予生理盐水。染毒组采用灌胃方式进行染毒,染毒剂量为0.01 mL/g,每天1次,母鼠受孕第1天至仔鼠出生后第19天灌胃染毒重铬酸钾,待仔鼠出生45 d后麻醉剖取卵巢。测定各组仔鼠体重、卵巢系数,以及卵巢组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力等。结果与对照组比较,低浓度染毒组仔鼠体重较高,而中、高浓度染毒组仔鼠体重明显较低,且以高浓度染毒组较为明显,不同浓度染毒组仔鼠卵巢重量、脏器系数、SOD活力,以及中、高浓度染毒组仔鼠GSH-Px活力均明显降低,差异有统计学意义(P<0.05);且随着重铬酸钾染毒浓度升高,仔鼠卵巢系数,以及SOD、GSH-Px活力均呈下降趋势,差异均有统计学意义(P<0.05)。结论母鼠孕期和哺乳期暴露重铬酸钾可致仔鼠卵巢组织氧化损伤。Objective To explore the effects of potassium dichromate exposure during pregnancy and lactation on ovarian damage in the filial rats.Methods In July 2019,a total of 15 seven-week-old female specific pathogen free SD rats with similar body weight,coat color and physiological status were divided into the low(1/8 LD50),medium(1/4 LD50),high(1/2 LD50)exposure groups(n=4)and control group(n=3).The control group was given normal saline.The exposed group was given intragastric administration at a dose of 0.01 mL/g,once a day.The potassium dichromate was administered intragastrically in the female rats from the first day of pregnancy to the 19th day after the filial rats′birth.The ovaries of filial rats were dissected under anesthesia 45 days after birth.The body weight,ovarian coefficient and the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in ovarian tissue of each group were measured.Results Compared with that in the control group,the body weight of the filial rats in the low exposure group was higher,while the body weight in the medium and high exposure groups were significantly lower,especially in the high exposure group.The ovary weight,organ coefficient,SOD activity and GSH-Px activity of the filial rats in different exposed groups decreased significantly,and the differences were statistically significant(P<0.05).With the increase of the potassium dichromate exposure,the ovary coefficient and the activity of SOD and GSH-Px decreased.The differences were statistically significant(P<0.05).Conclusion Exposure to potassium dichromate during the famle rats′pregnancy and lactation can cause oxidative damage to the ovary tissue of the filial rats.
