摘  要:AP2/ERF类转录因子是植物所特有的一类转录因子家族,在植物的生长发育及响应非生物胁迫中发挥着重要作用。为发掘桑树AP2/ERF类转录因子的成员及其功能,从桂桑优12中克隆到7个AP2/ERF转录因子,命名为MnAP2/ERF1~MnAP2/ERF7。生物信息学分析结果表明,这7个AP2/ERF蛋白都属于不稳定蛋白,且均属于亲水性蛋白,二级结构以无规则卷曲为主,亚细胞定位显示主要位于细胞核中。氨基酸序列比对及进化树分析发现,这7个AP2/ERF转录因子均含有1个保守AP2结构域,其中MnAP2/ERF1~MnAP2/ERF4属于ERF亚家族,MnAP2/ERF5~MnAP2/ERF7属于DREB亚家族。荧光定量分析显示,NaCl、PEG_(6000)、水淹胁迫可诱导7个桑树AP2/ERF基因上调表达,其中MnAP2/ERF3、MnAP2/ERF5、MnAP2/ERF6分别响应水淹、干旱和盐胁迫,说明桑树AP2/ERF转录因子的表达与逆境胁迫响应之间有密切的联系。本研究结果可为进一步揭示AP2/ERF转录因子调控桑树生长发育及逆境胁迫的机理奠定基础。The AP2/ERF transcription factors are a family of transcription factors unique to the plant,and they play important roles in plant growth and development and response to stresses.In order to discover members and functions of the AP2/ERF transcription factors of mulberry,seven AP2/ERF transcription factors were cloned from Guisang You 12 and named MnAP2/ERF1~MnAP2/ERF7.The results of bioinformatics analysis showed that the 7 proteins were all unstable proteins and hydrophilic proteins.The secondary structure of the proteins were mainly random coils,and the subcellular location showed that they were mainly located in nucleus.Amino acid sequence alignment and phylogenetic tree analysis revealed that all the 7 AP2/ERF transcription factors contained one conserved AP2 domain,among which,MnAP2/ERF1~MnAP2/ERF4 belonged to the ERF subfamily,while MnAP2/ERF5~MnAP2/ERF7 belonged to the DREB subfamily.Fluorescence quantitative analysis showed that the 7 AP2/ERF transcription factors expressed to varying degrees induced by salt,drought and flooding stresses.Among them,MnAP2/ERF3,MnAP2/ERF5 and MnAP2/ERF6 responded to flooding,drought and salt stresses respectively,indicating that the expression of mulberry AP2/ERFs had close relationships with the response to stresses.The results of this study could lay theoretical foundations for further revealing the mechanisms of AP2/ERF transcription factors regulating the growth and development of mulberry trees and response to abiotic stresses.
