‘翠鸟’玉簪组培快繁技术初探

摘　　要：[目的]找出‘翠鸟’玉簪外植体最佳灭菌方法,适合愈伤组织诱导、丛生芽增殖和生根的最佳激素配比的培养基配方。[方法]以"翠鸟"玉簪幼嫩的花葶作为外植体进行快繁研究。[结果]外植体最佳灭菌方法是氯化汞5 min,愈伤组织诱导的最佳培养基为MS+6-BA 2.0 m L/L+NAA 0.1~0.5 ml/L,最适合丛生芽增殖的组合为MS+6-BA 1.0 m L/L+NAA 0.01~0.1 m L/L,生根1/2 MS+NAA 0.01~0.05 m L/L为宜。[结论]‘翠鸟’玉簪组培生根苗移栽应严格注意环境控制。[Objective]The aim was to find out the best sterilization method of Hosta plantaginea’Halcyon’explants,which was suitable for callus induction,cluster bud proliferation and rooting medium formula of the best hormone ratio.[Method]The young flower-scape of Hosta plantaginea’Halcyon’was used as explant to study rapid propagation.[Result]The best sterilizing method for explants was mercury chloride for 5 min,the best medium for callus induction was MS+6-Ba 2.0 m L/L+NAA 0.1-0.5 m L/L,and the best combination of MS+6-BA 1.0 m L/L+NAA 0.01-0.1 m L/L.The suitable rooting condition was 1/2 MS+NAA 0.01-0.05 m L/L.[Conclusion]Strict attention should be paid to environmental control for the tissue culture rooting and transplanting of Hosta plantaginea’Halcyon’.
