摘  要:本试验旨在研究发酵麸皮多糖(FWBPs)对杜寒杂交肉羊瘤胃组织紧密连接蛋白和细胞因子基因表达及瘤胃菌群结构的影响。选取21只健康且体重[(21.85±0.64)kg]相近的6周龄杜寒杂交肉羊21只,随机分为3个组,每组7只羊。对照组、低剂量组和高剂量组分别在基础饲粮中添加0、200、400 mg/kg BW的FWBPs。预试期14 d,正试期56 d。结果表明:1)与对照组相比,低剂量组肉羊瘤胃组织中闭锁蛋白(occludin)、闭合蛋白-1(claudin-1)和闭合蛋白-4(claudin-4)的mRNA相对表达量显著增加(P<0.05),高剂量组肉羊瘤胃组织中occludin、claudin-1、claudin-4、闭合小环蛋白-1(ZO-1)、干扰素-γ(IFN-γ)和白细胞介素-6(IL-6)的mRNA相对表达量显著增加(P<0.05)。2)高剂量组肉羊瘤胃菌群Shannon指数和Simpson指数显著高于对照组和低剂量组(P<0.05)。与对照组相比,低剂量组肉羊瘤胃菌群中螺旋菌门(Spirochaetota)和琥珀酸弧菌属(Succinivibrio)的相对丰度显著增加(P<0.05);高剂量组肉羊瘤胃菌群中变形菌门(Proteobacteria)、琥珀酸弧菌科_UCG-001(Succinivibrionaceae_UCG-001)和解琥珀酸菌属(Succiniclasticum)的相对丰度显著或极显著降低(P<0.05或P<0.01),拟杆菌门(Bacteroidota)和新月形单胞菌属(Selenomonas)的相对丰度显著增加(P<0.05)。高剂量组肉羊瘤胃菌群中Proteobacteria、Spirochaetota和Succinivibrionaceae_UCG-001的相对丰度显著或极显著低于低剂量组(P<0.05或P<0.01),而Bacteroidota、韦荣球菌科_UCG-001(Veillonellaceae_UCG-001)和Selenomonas的相对丰度极显著高于低剂量组(P<0.01)。3)线性判别分析效应大小(LEfSe)分析显示,柔膜菌目(Erysipelotrichales)、氨基酸球菌科(Acidaminococcaceae)、氨基酸球菌目(Acidaminococcales)、Succiniclasticum在对照组中富集,琥珀酸弧菌科(Succinivibrionaceae)、气单胞菌目(Aeromonadales)、γ-变形菌纲(Gammaproteobacteria)、Proteobacteria、Succinivibrionaceae_UCG-001在低剂量组中富集,BacteThis study was conducted to investigate the effects of fermented wheat bran polysaccharides(FWBPs)on expression of tight junction protein and cytokine genes in rumen tissue and rumen microbiota structure of Dorper×thin-tailed Han crossbred meat lambs.Twenty-one 6-week-old Dorper×thin-tailed Han crossbred meat lambs with similar body weight[(21.85±0.64)kg]were randomly assigned into three groups of seven lambs in each group.Lambs in the three groups were fed basal diets supplemented with 0(control group),200(low dose group)and 400 mg/kg BW(high dose group)FWBPs,respectively.The pre-experimental period lasted for 14 days,and the experimental period lasted for 56 days.The results showed as follows:1)compared with the control group,the mRNA relative expression levels of occludin,claudin-1 and claudin-4 in rumen tissue of meat lambs of low dose group were significantly increased(P<0.05),and the mRNA relative expression levels of occludin,claudin-1,claudin-4,zonula occludens-1(ZO-1)interferon-γ(IFN-γ)and interleukin-6(IL-6)in rumen tissue of meat lambs of high dose group were significantly increased(P<0.05).2)The Shannon index and Simpson index in rumen microbiota of meat lambs of high dose group were significantly higher than those of the control and low dose group(P<0.01).Compared with the control group,the relative abundances of Spirochaetota and Succinivibrio in rumen microbiota of meat lambs of low dose group were significantly increased(P<0.05);the relative abundances of Proteobacteria,Succinivibrionaceae_UCG-001 and Succiniclasticum in rumen microbiota of meat lambs of high dose group were significantly decreased(P<0.05 or P<0.01),and the Bacteroidota and Selenomonas relative abundances were significantly increased(P<0.05).The relative abundances of Proteobacteria,Spirochaetota and Succinivibrionaceae_UCG-001 in rumen microbiota of meat lambs of high dose group were significantly lower than those of low dose group(P<0.05 or P<0.01),while the Bacteroidota,Veillonellaceae_UCG-001 and Selenomonas relative abun
