摘 要:目的探讨中药小分子单体β-桉叶醇对内皮细胞血管新生的影响及其分子作用机制。方法将小鼠脑微静脉内皮细胞分为空白组、DMSO组、β-桉叶醇组(n=3),分别采用0.1%磷酸盐缓冲液,0.1%DMSO,0.1%20 mg/L的β-桉叶醇药物溶液(溶剂为DMSO)的完全培养液培养。分析各组CD31、血管内皮生长因子A(VEGFA)、细胞周期蛋白D1(CyclinD1)、原癌基因C-Myc、基质金属蛋白酶(MMP)-2、MMP-9、增殖细胞核抗原(PCNA)、Bax、Bcl-2、半胱氨酸天冬氨酸蛋白酶(Caspase)-3、活化Caspase-3、P53、高迁移率族蛋白B1(HMGB1)蛋白表达。结果空白组与DMSO组各项指标比较,无统计学差异(P>0.05),与空白组及DMSO组比较,β-桉叶醇组细胞增殖能力、S期细胞比例、细胞周期相关蛋白C-Myc、CyclinD1、PCNA、MMP-2、MMP-9、CD31、VEGFA、HMGB1及Bcl-2表达显著下降(P<0.05),G1期细胞比例、细胞早期凋亡率、Bax、P53及活化Caspase-3表达上升[(73.73±1.70)%vs(61.33±3.04)%和(60.96±0.98)%,(26.33±2.67)%vs(9.31±0.52)%和(9.53±0.94)%,0.92±0.07 vs 0.33±0.05和0.32±0.02,0.50±0.07 vs 0.30±0.09和0.27±0.10,0.49±0.13 vs 0.14±0.08和0.13±0.11,P<0.05]。结论β-桉叶醇抑制内皮细胞血管新生,其机制可能是影响细胞增殖、迁移能力、调控细胞周期和凋亡相关信号通路。Objective To study the effect of β-eudesmol on angiogenesis of endothelial cells and its molecular mechanism.Methods The mouse bEnd.3 cells were divided into blank control group, DMSO group and β-eudesmol group.The bEnd.3 cells were washed in 0.1% BPS and cultured in 0.1% DMSO,0.1% β-eudesmol solution.The expressions of CD31,VEGFA,CyclinD1,C-Myc, MMP-2,MMP-9,PCNA,Bax, Bcl-2,Caspase-3,activated Caspase-3,and HMGB1 in different groups were detected and analyzed.Results No significant difference was detected in different parameters between blank control group and DMSO group(P>0.05).The proliferation rate of bEnd.3 cells, ratio of S phase bEnd.3 cells, and expression levels of C-Myc, CyclinD1,PCNA,MMP-2,MMP-9,CD31,VEGFA,HMGB1 and Bcl-2 were significantly lower in β-eudesmol group than in blank control group and DMSO group(P<0.05) while the rate of G1 bEnd.3 cells, ratio of early opoptosis of bEnd.3 cells, and the expression levels of Bax, P53 and activated Caspase-3 were significantly higher in β-eudesmol group than in blank control group and DMSO group(73.73%±1.70% vs 61.33%±3.04% and 60.96%±0.98%,26.33%±2.67% vs 9.31±0.52% and 9.53%±0.94%,0.92±0.07 vs 0.33±0.05 and 0.32±0.02,0.50±0.07 vs 0.30±0.09 and 0.27±0.10,0.49±0.13 vs 0.14±0.08 and 0.13±0.11,P<0.05).Conclusion β-eudesmol inhibits the angiogenesis of endothelial cells by affecting the proliferation and migration of bEnd.3 cells, and regulating the cell cycle and apoptosis-related signaling pathway.
