葛根素对妊娠期糖尿病大鼠的治疗效果及作用机制的研究

摘　　要：目的研究葛根素对妊娠期糖尿病大鼠的治疗效果及作用机制。方法妊娠SD大鼠随机分为正常妊娠组、妊娠期糖尿病组及葛根素组。妊娠SD大鼠腹腔注射链脲佐菌素建立GDM模型。葛根素组每日给予葛根素150 mg/kg灌胃治疗,糖尿病组与正常妊娠组每日给予等量的生理盐水。检测孕鼠血糖水平,妊娠第20天记录胎仔数、胎鼠体质量、畸形发生数、胎鼠血糖水平及孕鼠血浆胰岛素水平。苏木精-伊红染色观察大鼠胰腺组织结构改变情况,免疫组织化学及Western blotting检测葡萄糖转运蛋白-4(GLUT-4)的表达情况。结果葛根素可以显著降低GDM大鼠的血糖、增加胎仔数,降低胎鼠血脂和体质量。葛根素治疗能恢复胰岛数量和体积,同时增加GLUT-4蛋白表达。结论葛根素可以有效控制妊娠期糖尿病大鼠血糖水平,其作用机制可能与修复胰岛并上调胰岛内GLUT-4的表达有关。Objective:To study the therapeutic effects and mechanism of action of puerarin on gestational diabetes mellitus(GDM)in rats.Methods:All rats were randomly divided into 3 groups:the control group,the GDM group,and the puerarin group.GDM was induced in pregnant Sprague-Dawley rats by injection of streptozotocin.The rats in the puerarin group received puerarin gavage treatment(150 mg/kg),while the equal volume of normal saline was used for the control group and the GDM group.The random blood glucose was measured daily.Plasma glucose,serum insulin,body weight,and serum lipid were examined on 20 days of gestation.The number,weight,and blood glucose of fetuses were recorded.Hematoxylin and eosin staining showed pathological changes of pancreas.The expression of glucose transporter-4(GLUT-4)protein analyzed by immunohistochemistry and Western blotting.Results:Puerarin could reduce blood glucose level in GDM rats induced by streptozotocin.At the same time,it lowered body weight and serum lipid,and further improved serum insulin.Puerarin decreased the weight and blood glucose level of fetuses and increased litter size.Puerarin alleviated pancreatic pathological damage in GDM rats.Puerarin reduced hyperglycemia through recovering damaged islets,recovering insulin secretion,and upregulating GLUT4 expression in pancreas.Conclusion:Puerarin is an effective treatment for GDM in that it has protection on islet and improves the expression of GLUT-4 in pancreas.
