摘  要:目的探讨番石榴叶提取物对Zucker糖尿病肥胖(Zucker diabetic fatty,ZDF)大鼠胰腺组织microRNA表达水平的影响。方法实验共纳入12只6周龄雄性ZDF大鼠及6只同龄雄性Zucker瘦型(Zucker lean,ZL)大鼠,适应性喂养1周后,ZDF大鼠使用高脂饲料喂养,ZL大鼠使用普通饲料喂养。诱导喂养2周后,将6只ZL雄性大鼠作为正常组,12只ZDF大鼠检测随机血糖≥11.1 mmol/L者纳入实验,并随机分为模型组和治疗组,每组各6只大鼠。治疗组大鼠每日按0.01 mL/g体质量(折合生药材8 g/kg)灌胃给药,正常组、模型组大鼠灌服等体积蒸馏水,共干预4周。每周监测大鼠体重和空腹血糖。实验结束后,将大鼠心脏放血处死,并分离血清测定胰岛素水平,剖取胰腺并采用基因芯片检测模型组、治疗组大鼠胰腺中microRNA的表达。结果与模型组大鼠比较,治疗组大鼠空腹血糖和空腹胰岛素水平均降低,基因芯片检测到两组大鼠胰腺组织中共有9种microRNA差异性表达(7种上调,2种下调),其中miR-361-5p的表达显著上调。结论番石榴叶提取物干预后影响了大鼠胰腺中microRNA的表达,并可能通过调控miR-361-5p的表达调节胰岛β细胞去分化,从而降低血糖。Objective To explore the effect of guava leaves extracts on microRNA expression in pancreatic tissue of Zucker diabetic fatty(ZDF)rats.Methods A total of 12 six-weeks-old male ZDF rats and 6 male Zucker lean(ZL)rats of the same age were included in the experiment.After 1 week of adaptive feeding,the ZDF rats were given high-fat diet feeding and ZL rats were fed with ordinary feed.Then 6 ZL rats were regarded as the normal group,and 12 ZDF rats with blood glucose≥11.1 imnol/L were randomly divided into model group and treatment group.The rats of treatment group were given daily intragastric administration at 0.01 mL/g body weight(equivalent to 8 g/kg of crude medicinal materials)for 4 weeks.And other rats were given the same amount of distilled water.The body weight and fasting blood glucose of rats were monitored weekly.After the experiment,all rats were sacrificed and the blood was taken to determine the level of insulin,and the Agilent miRNA microarray was used to detect the expression of microRNA in pancreas of groups.Results Compared with the rats in model group,the fasting blood glucose and fasting serum insulin level of the rats in treatment group were significantly reduced.The microarray detected 9 types of microRNA differential expression in the pancreatic tissues.We found that 7 types were up-regulated and 2 types were down-regulated.Among them,the expression of miR-361-5p is up-regulated most obviously.Conclusion Guava leaves extracts affect the expression of microRNA in pancreas,and it may regulate pancreaticβ-cell dedifferentiation by miR-361-5p to decrease blood glucose.
