摘 要:目的:探究人SST抗独特型卵黄抗体对大鼠胰腺SST受体定位、荧光共定位及对胰腺内分泌功能的调节作用,探讨其能否用于糖尿病治疗。方法:DAB染色观察大鼠胰腺SST受体分布。免疫荧光共定位观察大鼠胰岛SST受体与内分泌激素共定位。大鼠腹腔注射SST抗独特型卵黄抗体,ELISA检测不同时间点大鼠血浆胰高血糖素、胰岛素和血糖水平。建立糖尿病大鼠模型,腹腔注射人SST抗独特型卵黄抗体,血糖仪检测治疗前后血糖水平。糖耐量实验检测糖尿病大鼠糖耐量水平。结果:DAB染色显示,SST受体免疫反应阳性物质除分布于胰岛细胞外,还分布于胰岛毛细血管内皮细胞和淋巴细胞。免疫荧光共定位显示,SST受体与生长抑素共定位于D细胞,与胰高血糖素共定位于A细胞,与胰岛素共定位于B细胞。ELISA显示注射SST抗独特型卵黄抗体后不同时间点,大鼠胰高血糖素水平逐渐下降(P<0.01)。胰岛素水平明显降低,30 min时差异有统计学意义(P<0.01)。不同时间点大鼠血糖水平逐渐降低(P<0.05)。糖尿病大鼠腹腔注射SST抗独特型卵黄抗体后,大鼠血糖水平逐渐降低,治疗后第5~10天差异有统计学意义(P<0.05)。糖耐量实验同样显示治疗组相比于不治疗组糖耐量明显改善(P<0.01)。结论:人SST抗独特型卵黄抗体可与SST受体不同亚型结合,并能模拟SST功能调节胰岛内分泌激素及血糖水平,对糖尿病大鼠有明显降血糖作用,提示其对糖尿病可能有治疗效果。Objective:To investigate effect of human SST anti-idiotypic yolk antibody on SST receptor localization,fluorescence co-localization and endocrine function regulation of rat pancreas,and to explore whether it could be used in treatment of diabetes.Methods:DAB staining was used to observe distribution of SST receptors in pancreas of rats. Immunofluorescence was used to observe co-localization of SST receptor and endocrine hormone in rat islet. Rats were intraperitoneally injected with SST anti-idiotypic yolk antibody and plasma glucagon,insulin and blood glucose levels were detected by ELISA at different time points. Diabetic model of rats were established and intraperitoneally injected by human SST anti-idiotypic yolk antibody. Blood glucose levels were detected by glucose instrument. Glucose tolerance level of diabetic rats was detected by glucose tolerance test.Results:DAB staining showed that SST receptor immunoreactive positive substances were not only distributed in islet cells but also in endothelial cells of blood capillary and lymphocytes. Immunofluorescence co-localization showed that SST receptor was co-localized in D cells with somatostatin,in B cells with insulin,in A cells with glucagon. ELISA showed that glucagon level was decreased gradually at different time points(P<0.01)after injection of SST anti-idiotypic yolk antibody. Insulin level was decreased significantly with statistical significance at 30 min(P<0.01). Blood glucose level of rats was also decreased gradually at different time points(P<0.05). After intraperitoneal injection of SST anti-idiotypic yolk antibody in diabetic rats,glucose was gradually decreased,which was significantly at 5~10 d(P<0.05). Glucose tolerance test also showed that glucose tolerance in treatment group was significantly improved than untreated group(P<0.01).Conclusion:Human SST anti-idiotypic yolk antibody can bind to different subtypes of SST receptors,and simulate function of SST to regulate islet endocrine hormone and blood glucose level in diabetic rats,signific
