健脾化瘀解毒方调控能量代谢抑制肺癌细胞侵袭转移的研究

摘　　要：目的:研究健脾化瘀解毒方的含药血清(JHJRMS)对人肺癌细胞能量代谢的影响。方法:TCGA数据库分析己糖激酶2(HK2)与肺癌患者预后之间的相关性;划痕实验用来检测JHJRMS对肺癌细胞侵袭转移的影响。RT-PCR检测JHJRMS对肺癌细胞中miR-143-3p表达水平的影响。数据库预测miR-143-3p与HK2蛋白的相关性。Western blot检测JHJRMS对肺癌细胞HK2、E-cadherin、MMP-2、MMP-9蛋白的影响。结果:TCGA数据库结果显示,HK2低表达,肺癌患者具有更好的预后。划痕实验结果显示JHJRMS能显著抑制肺癌细胞的侵袭转移。RT-PCR结果显示JHJRMS可明显增加肺癌细胞中miR-143-3p的表达水平。数据库结果提示,miR-143-3p和蛋白HK2的mRNA存在多处互补链,miR-143-3p可能靶向调控HK2。Western blot结果显示JHJRMS处理肺癌细胞后,细胞中的HK2、E-cadherin、MMP-2、MMP-9蛋白表达明显降低。结论:JHJRMS能明显抑制人肺癌A549细胞的侵袭转移,其机制可能是通过miR-143-3p调控HK2的表达水平直接影响细胞的能量代谢过程。Objective:To study the effect of Jianpi Huayu Jiedu recipe medicated serum(JHJRMS)on energy metabolism of human lung cancer cells.Methods:TCGA database analyzed the correlation between HK 2 and the prognosis of lung cancer patients.The scratch test was used to detect the influence of JHJRMS on the invasion and metastasis of lung cancer cells.RT-PCR was used to detect the effect of JHJRMS on the expression of miR-143-3p in lung cancer cells.The database predicted the correlation between miR-143-3 p and HK 2 protein.Western blot was used to detect the effects of JHJRMS on HK 2,E-cadherin,MMP-2 and MMP-9 proteins in lung cancer cells.Results:The results of TCGA database showed that patients with the lower expression of HK 2 had a better prognosis.The scratch test results showed that JHJRMS could significantly inhibit the invasion and metastasis of lung cancer cells.RT-PCR results showed that JHJRMS could significantly increase the expression of miR-143-3p in lung cancer cells.The database results suggested that there were multiple complementary strands in the mRNA of miR-143-3p and protein HK 2,and miR-143-3p was likely to target and regulate HK 2.Western blot results showed that after JHJRMS treated lung cancer cells,expressions of HK 2,E-cadherin,MMP-2 and MMP-9 proteins in the cells was significantly reduced.Conclusion:JHJRMS can significantly inhibit the invasion and metastasis of human lung cancer A 549 cells,which may be related to the regulation of HK 2 expression by miR-143-3p.
