摘 要:目的:探讨蔓荆子黄素对胃癌细胞增殖、凋亡的影响及可能机制。方法:不同浓度的蔓荆子黄素作用于胃癌细胞AGS后,CCK-8实验和流式细胞术分别检测细胞的增殖和凋亡,RT-qPCR检测miR-1193表达量,Western blot检测CyclinD1、p21、Bcl-2和Bax蛋白表达。将miR-1193模拟物转染至AGS细胞,检测上调miR-1193对AGS细胞增殖和凋亡的影响;将miR-1193抑制物转染至AGS细胞,检测下调miR-1193对蔓荆子黄素处理的AGS细胞增殖和凋亡的影响。结果:蔓荆子黄素作用后,AGS细胞的增殖能力下降,CyclinD1和Bcl-2表达降低,细胞凋亡率升高,miR-1193和Bax表达升高,且呈一定的剂量依赖效应(P<0.05)。上调miR-1193后AGS细胞增殖能力、CyclinD1和Bcl-2表达降低,凋亡率、Bax表达升高(P<0.05)。下调miR-1193表达能够逆转蔓荆子黄素对AGS细胞增殖和凋亡的影响(P<0.05)。结论:蔓荆子黄素能够抑制胃癌细胞增殖,诱导细胞凋亡,其机制可能与上调miR-1193表达有关。Objective:To explore the effect of vitexicarpin on proliferation and apoptosis of gastric cancer cells and its possible mechanism.Methods:Gastric cancer cell AGS was treated with different concentrations of vitexicarpin. CCK-8 assays and flow cytometry were used to detect cell proliferation and apoptosis,respectively. RT-qPCR was used to detect miR-1193 expression. Western blot was used to detect protein expressions of CyclinD1,p21,Bcl-2 and Bax. miR-1193 mimics was transfected into AGS cells,and effect of miR-1193 up-regulation on proliferation and apoptosis of AGS cells was detected by the above methods. miR-1193 inhibitor was transfected into AGS cells,and effects of miR-1193 down-regulation on proliferation and apoptosis of AGS cells treated with vitexicarpin were detected by the above methods.Results:After treated with vitexicarpin,Proliferation ability of AGS cell was decreased,CyclinD1 and Bcl-2 expressions were decreased,while apoptosis rate was increased,miR-1193 and Bax expressions were increased,and showing a certain dose-dependent effect(P<0.05). After up-regulation of miR-1193,AGS cell proliferation ability,CyclinD1 and Bcl-2 expressions decreased,while apoptosis rate and Bax expression were increased(P<0.05). Down-regulation of miR-1193 expression could reverse the effects of vitexicarpin on proliferation and apoptosis of AGS cells(P<0.05).Conclusion:Vitexicarpin can inhibit proliferation and induce apoptosis of gastric cancer cells,and its mechanism may related to the up-regulation of miR-1193 expression.
