摘  要:目的探讨蛋氨酸负荷对去卵巢大鼠氧化应激的改变及叶酸对其影响。方法4月龄雌性SD大鼠32只随机分为4组:假手术组(SHAM组)、去卵巢组(OVX组)、蛋氨酸负荷组(去卵巢+蛋氨酸,MET组)、叶酸治疗组(去卵巢+蛋氨酸+叶酸,FA组)。去卵巢术后1周,给予蛋氨酸灌胃及行叶酸干预治疗9周后,测定各组血清同型半胱氨酸(Hcy)、总抗氧化能力(TAC)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)的变化。结果MET组血清TAC、GSH-Px活性较SHAM组、OVX组明显下降,而Hcy、MDA浓度显著升高(P<0.05),FA组TAC、GSH-Px活性较OVX组、MET组显著升高(P<0.05),Hcy、MDA浓度明显下降。结论在去卵巢的情况下,叶酸作为氧自由基清除剂可改善高同型半胱氨酸血症引起的氧化应激损伤。Objective To investigate the effect of methionine load on oxidative stress and folic acid in ovariectomized rats.Methods 4-month-old female SD rats(n=32)were randomly divided into four groups:sham operation group(SHAM group),ovariectomized group(OVX group),methionine group(OVX+methionine,MET group)and folic acid treatment group(ovariectomized+methionine+folic acid,FA group).After one week of ovariectomy and 9 weeks of intervention with oral methionine,or folate line,serum homocysteine(Hcy),total antioxidant capacity(TAC),glutathione peroxidase(GSH-Px),propylene dialdehyde(MDA)were determined.Results The activity of serum TAC,GSH-Px in MET group was lower than those in SHAM,OVX group,while the concentration of Hcy,MDA was significantly higher in MET group(P<0.05).The activity of TAC,GSH-Px in FA group was higher than those in OVX,MET group,while the concentration of Hcy,MDA were decreased significantly.Conclusion Folic acid as oxygen free radical scavengers can improve the oxidative stress induced by hyperhomocysteine under the condition of ovariectomization.
