摘 要:目的探究磁性活性细胞分选法(MACS)、密度梯度离心法(DGC)、上游法(SU)、洗涤法(SW)四种精液处理方法在冷冻精子优选中的应用价值。方法41例志愿者的精液样本为研究对象,将其等分为4份,制备冷冻精液后,分别应用MACS、DGC、SU、SW法处理,分别设为A组、B组、C组、D组。比较四组优选前后的精子总数、向前运动精子活率、精子形态学状态。结果A、B、C、D组优选后的精子总数分别为(38.97±10.98)×10^(6)/ml、(22.30±6.96)×10^(6)/ml、(10.45±2.33)×10^(6)/ml、(37.12±12.49)×10^(6)/ml,均低于本组优选前,且C组的精子总数低于A、B、D组,B组的精子总数低于A、D组,差异具有统计学意义(P<0.05)。优化后,A、D组的向前运动精子活率分别为(22.74±6.88)%、(28.79±7.20)%,低于本组优选前,B、C组的向前运动精子活率分别为(78.47±11.26)%、(82.37±21.00)%,高于本组优选前,且B、C组的向前运动精子活率高于A、D组,D组的向前运动精子活率高于A组,差异具有统计学意义(P<0.05)。A、B、C、D组优选后的精子质膜完整率分别为(84.77±11.45)%、(81.30±6.48)%、(81.94±10.34)%、(71.11±6.71)%,精子DNA完整率分别为(91.13±3.67)%、(84.56±7.44)%、(85.36±8.82)%、(79.30±5.15)%,均高于本组优选前,且A、B、C组的精子质膜完整率、精子DNA完整率均高于D组,A组的精子DNA完整率高于B、C组,差异具有统计学意义(P<0.05)。结论在冷冻精子优选中,MACS、DGC、SU、SW方法各具优势,可依据临床实际需求选择优选方式。Objective To investigate the application of magnetic activated cell sorting(MACS),density gradient centrifugation(DGC),swim-up(SU)and simple washing(SW)in the selection of frozen sperm.Methods The semen samples of 41 volunteers were studied,and they were aliquoted into 4 portions,and after preparing frozen semen,they were treated by MACS,DGC,SU and SW methods respectively,and were set up as group A,group B,group C and group D,respectively.The total number of sperm,motility rate of progressive motility sperm,and state of sperm morphology before and after selection of the four groups were compared.Results The total number of sperm in groups A,B,C and D after selection were(38.97±10.98)×10^(6)/ml,(22.30±6.96)×10^(6)/ml,(10.45±2.33)×10^(6)/ml,and(37.12±12.49)×10^(6)/ml,which were lower than those before selection in this group;the total number of sperm in group C was lower than that in groups A,B,and D,and the group B was lower than groups A and D;all differences were statistically significant(P<0.05).After selection,the motility rate of progressive motility sperm of groups A and D were(22.74±6.88)%and(28.79±7.20)%,which were lower than those of this group before selection;the motility rate of progressive motility sperm in groups B and C were(78.47±11.26)%and(82.37±21.00)%,which were higher than those of this group before selection,and the groups B and C were higher than groups A and C,and the group D was higher than group A;all differences were statistically significant(P<0.05).After selection,the sperm membrane integrity rate in groups A,B,C,and D were(84.77±11.45)%,(81.30±6.48)%,(81.94±10.34)%,(71.11±6.71)%,and the sperm DNA integrity rate were(91.13±3.67)%,(84.56±7.44)%,(85.36±8.82)%,(79.30±5.15)%,which were all higher than those of this group before selection;the sperm membrane integrity rate and sperm DNA integrity rate of groups A,B,and C were higher than those of group D,and the sperm DNA integrity rate of group A was higher than that of groups B and C;all differences were statistically si
