摘 要:目的分析COVID-19病例咽拭子与鼻拭子标本新型冠状病毒(novel coronavirus 2019,2019-nCoV)核酸检测效果。方法对541例珠海市疑似病例和密切接触者,同时采集咽拭子和鼻拭子标本,用实时荧光PCR技术检测2019-nCoV核酸,分析两类标本核酸检出阳性率和靶标ORF1ab和N基因Ct值。结果共43例核酸确诊阳性,咽拭子共检测出35例阳性,阳性率6.47%,鼻拭子共检出33例阳性,阳性率6.10%,阳性率差异无统计学意义(χ^(2)=0.06,P=0.80),诊断一致性较好(Kappa=0.72)。两类标本ORF1ab和N基因的Ct值无差异。结论COVID-19病例鼻拭子和咽拭子标本采用实时荧光PCR技术检测2019-nCoV病毒核酸效果无差别。Objective To analyze the detection effect of novel coronavirus 2019 nucleic acid in throat swabs and nasal swabs of COVID-19 cases.Methods Throat swabs and nasal swabs were simultaneously collected from 541 cases of suspected cases and close contacts.Real time PCR was used to detect novel coronavirus 2019 nucleic acid.The positive rate of nucleic acid and Ct values of ORF1ab gene and N gene were analyzed.Results A total of 43 cases were confirmed positive by nucleic acid detection result;35 cases were positive and the positive rate was 6.47%in throat swabs,while 33 cases were positive and the positive rate was 6.10%in nasal swabs.There was no significant difference in the positive rate between the two types of samples(χ^(2)=0.06,P=0.80).The diagnostic consistency of the two specimens was good(Kappa=0.72).Ct values of ORF1ab and N genes between the two groups were not significantly different,either.Conclusions There was no significant difference in the detection of novel coronavirus 2019 between throat swabs and nasal swabs from COVID-19 patients by real-time PCR.
